[Objective] To observe the effects of Xintongfang on arrhythmia,infarct size,myocardial histopathology and CaMKⅡ gene expression in rats with acute myocardial infarction,and to explore the mechanism of Xintongfang on rats with acute myocardial infarction.[Methods] Fifty SD rats were randomly divided into five groups:sham-operated group,model group,Xintongfang high-dose group,Xintongfang low-dose group and Thiazolone group,rat models of acute myocardial infarction were established by ligating coronary artery,the levels of CK-MB,LDH and cTnT were detected by automatic biochemical analyzer,arrhythmia was detected by electrocardiogram,infarct size was measured by HE staining,morphological changes of myocardial cells were observed,and the expression of CaMKⅡ in myocardial tissue was determined by RT-PCR.[Results] There was no significant difference in the incidence of arrhythmia between Xintongfang high dose group and thiazephone group(P>0.05),Compared with Xintongfang low dose group,the incidence of arrhythmia was significantly lower(P< 0.05);The ischemic area of myocardial infarction in the high dose group of Xintongfang was significantly smaller than that in the thiazephone group and the low dose group of Xintongfang(P<0.01 or P<0.05);Xintongfang high dose group can reduce myocardial injury;Xintongfang high dose group inhibits the expression of CaMKⅡ mRNA more advantageous than thiazephone and Xintongfang low dose group(P<0.01).[Conclusion] Xintongfang high dose group has protective effect on acute myocardial infarction rats,its mechanism may be related to down-regulation of CaMK II gene expression in acute myocardial infarction rats.
