Objective:To investigate the relationship between the BRAF~V600E600E mutation and the express-ion of MMP-2 and MMP-9 in the papillary thyroid carcinoma.Methods:104 cases of papillary thyroid cancer tissues(44 cases of BRAF~V600E600E genotype of wild VS 60 cases of BRAF~V600E600E mutation type)and 20 cases of normal thyroid tissues in our hospital from January 2016 to June 2017 were selected as the research objects.Immunohistochemistry was used to detect the expression of MMP-2 and MMP-9 in papillary thyroid cancer tissues and normal thyroid tissues.The chisqare tests or Fisher were not only used to exmine the difference of the protein expression of MMP-2 and MMP-9 between PTC tissues and normal tissues,but also to analyze the correlation between their expression and pathologic characteristics.We applied qRT-PCR to test m RNA expression of MMP-2 and MMP-9 between PTC tissues(20 cases of BRAF~V600E600E genotype of wild VS 20 cases of BRAF~V600E600E mutation type)and normal paracancerous tissues.SPSS 23.0 statistical software was applied to analyze the relationship between the expression of MMP-2 and MMP-9 and BRAF~V600E600E mutation.Results:The protein and m RNA expression of MMP-2 and MMP-9 in PTC are significantly higher than in the normal tissues.The protein and mRNA expression of MMP-2 and MMP-9 in patients with BRAF~V600E600E mutation are significantly higher than those who don’t have.Analyses showed that the protein expression of MMP-2 and MMP-9 were significantly varied with capsular invasion,TNM stage and lymph node metastasis,which is not associated with age,gender and mulifocality.Conclusions:The protein and mRNA expression level of MMP-2 and MMP-9 increases in PTC tissues with BRAF~V600E600E mutation.The expression of MMP-2 and MMP9 in PTC is related to capsular invasion,TNM stage and lymph node metastasis,suggesting that high expression of MMP-2 and MMP-9 may be involved in the higher possibility of invasive metastasis with BRAF~V600E600E mutation.
