Objective:Lactate dehydrogenase(LDH)and urinary β2-microglobulin are both important clinical medical indicators.Lactate dehydrogenase is sensitive to the detection of various cancers, and urinary β2-microglobulin is of great significance for many diseases of kidney organs. The establishment of scientific reference value is the basis of clinical application of medical indicators. However, there is still some controversy about the reference value of lactate dehydrogenase and urinaryβ2-microglobulin in clinic.Since 1950s,a lot of researches had been down to the scope of health human‘s lactate dehydrogenase reference value, but the conclusions are different;and many scholars have studied the range of different areas of healthy human urine β2-microglobulin reference value, and the study showed that the differences ofregional differences will lead to urine β2-microglobulin reference value. At the same time, previous studies have shown that natural environmental factors such as altitude will affect the value of these two medical indicators.Based on the above problems, this paper selects two indexes of adult——lactate dehydrogenase and urineβ2-microglobulin to study. We research the geographical environment and its mechanism on these two medical indicators, establish prediction models of geographic factors and the medical indicators, develop the reference values of lactate dehydrogenase and urine β2-microglobulin in different regions and provide scientific reference value support for clinical diagnosis.Method:In this study,we retrieved medical literature and collected 10118 lactate dehydrogenase and 10313 urine β2-microglobulin values in healthy adults. In order to fully reflect the impact of geographical environment on medical indicators, this study selected 25 geographical factors from the spatial location, topography, climate and soil from four aspects, including longitude, latitude, altitude, annual sunshine hours, annual average, the annual average temperature, the annual average relative humidity, annual precipitation, annual temperature range, the annual average wind speed, the percentage of gravel soil, soil particle percentage, the percentage of soil clay, surface reference capacity, topsoil the bulk density, soil gravel content, soil organic matter content,pH,clay soil cation exchange capacity, soil cation exchange capacity, soil base saturation, soil total The amount of exchange, the content of calcium carbonate in the topsoil, the content of the topsoil calcium sulfate, the base degree of the topsoil and the salt of the surface soil.First of all, this study use correlation analysis to determine the main geographical influencing factors of lactate dehydrogenase and urine β2-microglobulin reference value;secondly, prediction models between the two medical indicators and relevant geographical factors were established by partial least squares model and trend surface model;then we contracted the model accuracy and the optimum prediction models of lactate dehydrogenase and urine β2-microglobulin were selected;thirdly,we calculated the reference value in China 2322 points according to the optimal prediction model, and then used Kriging interpolation and Hot spot analysis to research the spatial distribution characteristics of the reference values of these two medical indicators;finally, the effects of geographical environment on lactate dehydrogenase and urineβ2-microglobulin were analyzed according to the results.Result:The main geographic reference value of lactate dehydrogenase in healthy adults the impact factor of altitude, annual average relative humidity, soil sand percentage, topsoil particle percentage, soil clay percentage, soil bulk density, soil reference capacity, soil cation exchange capacity and total amount of soil exchangeable, correlation coefficients were 0.248,0.186,0.174,-0.163,-0.288-0.262,-0.164,0.290 and 0.158;the main geographic health adult urine β2-microglobulin reference value affecting factors:altitude, annual sunshine hours, annual average relative humidity, annual precipitation of clay and cation exchange capacity, the correlation coefficients were 0.231,0.268,-0.302,-0.166,0.186.The model verification results showed that the optimal prediction model for the reference value of lactate dehydrogenase and urineβ2-microglobulin was partial least square model. In spatial analysis, both lactate dehydrogenase and urine β2-microglobulin showed strong spatial distribution characteristics. Conclusion:The reference value of lactate dehydrogenase and the reference value of urine β2-microglobulin can be significantly affected by the geographical environment. This study made a comprehensive analysis and modeling for the relationship between the reference values of these two indicators and geographical environment. The research results can help to improve the accuracy of clinical diagnosis.
