Background:Long noncoding RNAs(IncRNAs),which comprise a class of non-coding RNAs(more than 200 nucleotides)without protein coding potential generated by RNA polymerase Ⅱ,are involved in various biological processes,including cell proliferation,differentiation,apoptosis,development and immune responses。Some recent reports have demonstrated that lncRNAs play critical roles in cancer development and progression and they have the potential to serve as diagnostic and prognostic biomarkers in gastric cancer,prostate cancer,and hepatocellular carcinoma,among other cancers.LncRNAs are tumor-specifc and are also very stable in plasma,urine and other easily obtainable body fluid,which indicates that they are good candidate tumor biomarkers.However,conclusions about the diagnostic value of lncRNAs in thyroid cancer are little known.Studies have shown that lncRNA NONHSAT037832 expressed significantly lower in papillary thyroid cancer tissues than adjacent noncancerous tissues.Objective:In this study,we will investigate the expression levels of long non-coding RNA NONHSAT037832(LncRNA NONHSAT037832)in tissues and plasma of patients with papillary thyroid cancer(PTC),and evaluate the diagnostic value of LncRNA NONHSAT037832 in PTC.Methods:1.Patients with PTC and thyroid adenoma who accepted operation were involved in our study.2.Patients were divided into three groups:benign thyroid adenoma,PTC without cervical lymph node metastasis(NO),PTC with cervical lymph node metastasis(N1).3.Real-time fluorescence quantitative polymerase chain reaction(RT-PCR)was applied to detect the expression levels of LncRNA NONHSAT037832 in patients’plasma before surgery and in paients’ tumor tissues.4.Receiver operating characteristic(ROC)curve was performed to evaluate the diagnostic value of LncRNA NONHSAT037832 in PTC.Results:1.The relative expression level of NONHSAT037832 in PTC and thyroid adenoma tissues was respectively 2.81±0.63 and 3.04±0.56 and the difference had no statistical significance(P>0.05);Similarly,the expression level of LncRNA NONHSAT037832 in patients’ plasma also had no significant difference(P>0.05).2.The relative expression level of NONHSAT037832 in PTC not accompanied by cervical lymph node metastasis and PTC with cervical lymph node metastasis was respectively 2.42±0.38 and 8.74±1.5,and the difference had evidently statistical significance(P<0.01);Similarly,the expression level of LncRNA NONHSAT037832 in patients’ plasma also had no significant difference(P>0.05);Identically,the expression level of plasma-derived LncRNA NONHSAT037832 in N1 group was obviously lower than that in NO group.3.A receiver operating characteristic(ROC)curve was established to determine the diagnostic significance of a NONHSAT037832 value for PTC with or without cervical lymph node metastasis.The area under the curve(AUC)was up to 0.882(95%CI:0.852-0.960,P<0.001).Conclusion:Expression level of LncRNA NONHSAT037832 in PTC tissues were positively correlated with that in PTC patients’ blood and LncRNA NONHSAT037832 level was of great diagnoatic value to discriminate PTC whether with cervical lymph node metastasis or not,suggesting that plasma-derived NONHSAT037832 will probably be the potential biomarker of PTC patients and provide an efficient guidance for the operation.
