Photodynamic therapy(PDT)as a new method for the treatment of cancer.This method Compared with traditional cancer treatment method has many advantages.A growing number of phthalocyanine were used as photosensitizer application of PDT.While previous studies have focused on phthalocyanine surrounding,axial modification is relatively less.Surrounding modification comparatively easy lead to aggregate.Agregate to cause a decline in phthalocyanine photosensitive activity.Axial modification compared to surrounding modification in terms of advantage after introducing suitable axial ligands,can prevent phthalocyanine ring of π-π aggregate.So this kind of phenomenon is largely improved the phthalocyanine photodynamic therapy.Based on this,this paper synthesized three axial replace silicon phthalocyanine,its structure is characterized and their properties have been pilot studied.The results of study are as follow.(1)With silicon phthalocyanino dichloro as raw material,three axial replace silicon phthalocyanine was synthesized compounds:SiPc1,SiPc2,SiPc3.All of the phthalocyanine compounds were characterized by IR spectra,UV-Vis spectra,1H-NMR,13C-NMR,MS.(2)By uv-vis absorption spectroscopy results shows that three axial replace silicon phthalocyanine respectively in water,DMF and cell lysis liquid solvents showed the characteristics of low aggregated.By fluorescence spectrum results shows that three axial replace silicon phthalocyanine,SiPcl and SiPc3 show high fluorescence intensity,and SiPc2 fluorescence intensity is low.(3)Light stability experiment showed that SiPc2 and SiPc3 has good stability in water.Singlet oxygen generation ability experiments show SiPcl and SiPc3 has higher yield of singlet oxygen in the water,in a physiological environment is also.So these compounds have a good potential of photosensitizer. |
