Objective: To observe the effects of polydatin on neurological deficits and neuronal apoptosis in cerebral ischemia / reperfusion injury(CIRI),the changes of Caspase-8and FLIP protein after cerebral ischemia / reperfusion injury,and the effect of polydatin to explore the protective mechanism of polydatin on cerebral ischemia /reperfusion injury.Methods:Ninety-six healthy male SD rats of SPF grade were randomly divided into three groups: sham operation group,model group and polydatin model group.The rats in the sham operation group and the model group were treated with normal saline10ml/(kg.d)every day.The polydatin model group was given paclitaxel 15mg/(kg.d)every day.Day gavage 1 time.The rats were sacrificed at 6h and 72 h after the completion of the model.The rats were sacrificed at 6h and 72 h after the completion of the model.The Longa method was used to evaluate the neurological function score.The The expression of Caspase-8 and FLIP protein in hippocampus of rats were detected by immunohistochemical staining.The expression of Caspase-8 and FLIP protein in hippocampus of rats were detected by RT-PCR.The expression of Caspase-8 and FLIP protein in hippocampus of rats were observed by TUNEL method and mRNA expression.SPSS17.0 software was used for statistical processing.Result:1.There were no significant differences in neurological deficits between the model group and the polydatin group(P> 0.05).There was no significant difference between the two groups at 6h and 72h(P> 0.05).Compared with the sham operation group,the model(P<0.05).Compared with the model group,the symptoms of nerve defects were decreased at different time points(P<0.05),and the difference was statistically significant(P <0.05).2.Apoptotic cells were observed in the sham operation group.Apoptotic cells were observed in the hippocampus and its surrounding ischemic side at 6h and 72 h after model group and paclitaxel group.Apoptotic cells were increased in the model groupcompared with the sham group(P<0.01).The expression of polydatin in the two groups was significantly higher than that in the model group(P<0.05).The expression of the apoptotic cells in the two groups was higher than that in the model group The difference between the two groups was statistically significant(P<0.05).3.The expression of Caspase-8 protein and mRNA in model group and paclitaxel group was significantly higher than that in sham operation group(P<0.05).Compared with sham operation group,the expression of Caspase-8 protein and mRNA in model group was significantly higher than that in sham operation group(P<0.01).Compared with the model group,the expression of Caspase-8 in the polydatin group was lower than that in the model group(P<0.05).The expression of Caspase-8 in the model group was significantly higher than that in the model group(P<0.01).There was no significant difference between the two groups(P>0.05).There was no significant difference between the two groups(P> 0.05).4.The FLIP protein and mRNA expression were not found in the sham operation group.The expression of FLIP protein and mRNA in model group and paclitaxel group increased at 6h and 72 h.Compared with the sham operation group,the expression of FLIP in the model group increased at different time points(P<0.05).The expression of FLIP in polydatin group was higher than that in model group(P<0.05).The expression of FLIP in polydatin group was higher than that in model group(P<0.05),and there was significant difference between the two groups(P<0.05).Conclusion:1.The expression of Caspase-8 and FLIP protein was significantly higher than that of the control group after cerebral ischemia / reperfusion injury in rats.2.Polydatin may promote the expression of FLIP by down-regulating Caspase-8,thereby reducing neuronal apoptosis and reducing brain injury after cerebral ischemia/ reperfusion injury.
