The Effect Of Electroacupuncture On The Discharge Of ACC Neurons And The Expression Of TRPV1 In The Colon Of IBS Model Rats

ObjectiveThis trail tried to observe the IBS model rats’ intestine sensitivity of pain reaction,emotional reaction and the expression of the change of TRPV1,discuss the different affection to the model rats’ corresponding symptoms,inspect the change of characteristic of neuron discharge in the region of ACC of the brain and analyze part of neural mechanism of the specific of acu-points affection through electro-acupuncture on PC6 and ST25.MethodsWistar rats were randomly divided into blank group(BG),model group(MG),neiguan(PC6)electro-acupuncture group(NG)and tianshu(ST25)electro-acupuncture group(SG).Except the BG,the other groups’ newborn rats were received colonic irritation by acetic acid,separated from their mother from postnatal day 2 to day 15 and given intestinalstimulation by dilated balloon three times at 6 week old.The rats were given implanted electrodes randomly and guarantee 3 rat were succeed at least in every group.After 1 week’s recovery,NG and SG accepted electro-acupuncture once every other day for 5 times,the BG and MG were not given any treatment.To observe the intestine sensitivity of pain reaction through AWR method,analyze the emotional reaction through Open-field test,inspect the expression change of TRPV1 by immunohistochemical technique and survey the rule of neuron discharge in ACC region in the brain by multi-channel in vivo recording technique.Results1.The different effect of the IBM rats’ intestine sensitivity on different acu-point.Comparing with the BG,the MG shorten the latency of pain,increased the number of contraction wave(p<0.01);comparing with the MG,the NG prolonged the latency of pain(p<0.05),the TG prolonged the latency of pain and decreased the number of contraction wave significantly(p<0.01);comparing with NG,the TG prolonged the latency of pain significantly(p<0.01).2.The emotional change of the IBM model rat on different acu-point.Comparing with the BG,the scores in horizontal motion and vertical motion of the MG were low(p<0.01);comparing with MG,the scores in horizontal motion of NG were increased(p<0.05).3.The expression change of TRPV1 in the plasma mucosa in the IBS model rat through electro-acupunctureComparing with BG,the expression of TRPV1 was increased in MG(p<0.01);comparing with MG,the expression was decreased in NG and TG(p<0.01),and there was significant difference.4.The effect to the characteristic of neuron discharge in the region of ACC of the brain through electro-acupuncture on PC6 and ST25.1)Comparing with BG,the frequency of A neuron discharge in ACC was higher in MG(p<0.05),Coeff.Var.ISI was changed in MG(p<0.05);comparing with MG,the frequency of A neuron discharge in AC C was lower in NG(p<0.01);Comparing with NG,the frequency of A neuron discharge in ACC was higher in TG(p<0.05).2)Comparing with BG,the frequency of B neuron discharge in ACC was lower in MG(p<0.05),Coeff.Var.ISI was decreased in MG(p<0.05);comparing with MG,the frequency of B neuron discharge in ACC was higher in TG(p<0.01),Coeff.Var.ISI was decreased in TG(p<0.05).ConclusionThe manifestation of model rats’ intestine sensitivity of pain reaction and emotional reaction is abnormal that is accord with the characteristics of IBS,which is to say that separate the rat from their mother,colonic irritation by acetic acid and intestinal stimulation by dilated balloon can make the IBS model.Electro-acupuncture on PC6 and ST25 can improve the IBM model rats’ intestine sensitivity of pain reaction and emotional psychological abnormalities in some degree,which can also decrease the expression of TRPV1 in the plasma mucosa and change the regular of neuron discharge in the region of ACC in the brain of the IBM model rat.The effect of rats’ intestine sensitivity of pain reaction and emotional psychological abnormalities through electro-acupuncture on PC6 and ST25 is different,PC6 of the hand-jueyin pericardium meridian have a special effect to change the emotional psychological abnormalities;ST25 of the foot-yangming stomach meridian have an obvious effect on intestinal function.There is a specific therapeutic effect by electro-acupuncture on PC6 and ST25 to treat IBS.
