| Osmanthus fragrans Lour is one of the top ten traditional flowers in China.In this study,the propagation system of‘Boyejingui’was further optimized from the aspects of sexual reproduction,asexual propagation and tissue culture.At the same time,the growth and development morphology,physiology and mineral element enrichment of Osmanthus fragrans were compared andanalyzed by environmental factors of different cultivation sites,which provided reference for propagation and Cultivation of Osmanthus fragrans.The main results are as follows:1.Soaking the seeds of Cinnamomum undulatum in 150 mg/L ABT-1 rooting powder for 4 hours at 50℃can greatly improve the germination rate,which is 54%.2.When cutting,the middle and upper part of the tree body with full peripheral buds,vigorous growth and no pests and diseases were selected as cuttings.4 river sand+3 rotten leaf soil+3 perlite were used as the substrate.The outdoor temperature should not be too high when cutting,and the high temperature will affect the survival rate of cutting to a certain extent.150 mg/L abt-1 can significantly improve the survival rate of cuttings and the number of roots,the survival rate reached 80%.At the same time,the addition of phosphate bacteria and abt-1 rooting powder can further improve the cutting effect.Add 150 ml of 10~6cfu/ml root phosphate bacteria to 1 l soil sample.For‘Boyejingui’cassia seedlings,the concentration of dephosphorizing bacteria in 10~9cfu/ml still maintained a considerable role in promoting,compared with the control,the plant height increased by 14%,and the root was significantly more expanded,the number of branches was also more.But this concentration is not suitable for cutting propagation of cinnamon,on the contrary,it will play a certain role in inhibition.3.In tissue culture of‘Boyejingui’auricula,LMC medium was selected for the primary culture of embryo.The added hormone was 0.2 mg/L Ba,pH was 5.8-6.0,the temperature of culture room was 24±2℃,the light intensity was 3000 LX,and the medium was changed every 7 days for 14 hours.The germination rate could reach 85.57%.4.In Wanzhou,Wuxi and Enshi cultivation sites,the content of common heavy metals in the soil did not exceed the national standard,the magnesium content in Wanzhou and Wuxi cultivation sites was significantly higher than that in Enshi cultivation sites;the potassium content in Wanzhou cultivation sites was significantly higher than that in Wuxi and Enshi,while the calcium content was significantly lower than the other two sites.In Wanzhou,the temperature reached 38℃,while the highest temperature in Wuxi was no more than 33℃,which was very beneficial to the growth of flower buds.The annual yield of flowers in Wuxi was significantly higher than that in Wanzhou and Enshi,and the mineral elements,β-carotene and flavone content of flowers were higher.The optimum temperature for the growth of‘Boyejingui’undulatum is 15-28℃.Strong sunshine and shade are not good for its growth,but also affect the rooting of cuttings.Wuxi is a relatively suitable cultivation area for‘Boyejingui’auriculatum in Northeast Chongqing. |
