In recent years,super-resolution microscope imaging technology has developed very rapidly.It has become one of the most commonly used optical super-resolution technologies at present due to its advantages of low excitation power,no special requirements for samples.Based on this,we have carried out relevant research on the structured light transmission sheet and image reconstruction algorithm for structured light illumination super-resolution fluorescence microscopy imaging.This thesis analyzes the current research status of structured light illumination super-resolution fluorescence microscopy system.Subsequently,5 new structured light transmission sheets are designed by ray tracing method,and it is concluded that semicircular line,elliptical line,cosine line,hyperbolic line and parabolic structured light transmission sheets can all produce cosine structured light stripes.Among them,stripes generated by elliptical lines are due to the stripes generated by other linear structured light transmitting sheets;At this time,height of the elliptical line structure light transmitting sheet on the top of the column is 1.36 and the thickness of the substrate is 2.58.Cosine structure light stripes with a contrast of 0.54 can be obtained.Afterwards,the paper makes theoretical calculation and ZEMAX simulation of the transversely propagating structured light transmission sheet,points out that the structured light transmission sheet can realize periodic structured light stripes,and gives the specific calculation formula of the transmittance of the upper surface of the transmission sheet.Finally,a joint image reconstruction algorithm is proposed.The algorithm combines the three-phase imaging in the first direction with two-phase imaging in the subsequent direction,and can reconstruct the super-resolution microscopic images in N directions only by using 2N + 1 attached frequency domain images.Furthermore,the algorithm has good noise suppression capability.Finally,the paper also carries out computer simulation experiments by combining the algorithm with several structural light transmission sheet parameters designed in the paper.The results show that the illumination light field generated by several structural light transmission sheets can achieve a resolution of 200 nm,and the elliptical line structural light transmission sheet has the best imaging capability.In general,this thesis designs six kinds of structured light transmission sheets and a super-resolution image reconstruction algorithm for structured light illumination,which can effectively suppress noise.
