| Objective:This article mainly observes and compares the effects of Jiaotai Pill and Coptidis on chronic low-grade inflammation in obese mice.Methods:In this experiment,C57BL/6J mice were randomly divided into normal group and high-fat group.The normal group was fed with normal rats,the high-fat group was given high-fat diet,and the high-fat group was randomly divided into mice.The model group(M),Jiaotai Pill group(JT),Coptidis group(HL)and Bifidobacterium group(PF).The model group continued to feed with high-fat diet,and the Jiaotai Pill Group,the Coptidis Group and the Bifidobacterium Group(Bifidobacterium Triple Active Bacterial Preparation Group)were treated with Jiaotai Pill Decoction and Coptidis at the same time as high-fat feeding.Bifidobacterium was treated with gavage for 4 weeks,fasting blood glucose(FBG)and glucose tolerance(IPGTT)were detected,and then the animals were sacrificed.Collect blood to detect fasting blood glucose(FBG);collect serum to detect endotoxin(LPS),insulin(INS),blood lipids(TG,TC,LDL-c,HDL-c);detect liver tissue TG,TC;ELISA The levels of TNF-α and IL-6 in liver tissue and the levels of MCP-1,TNF-α and IL-6 in epididymal fat were detected.The expression levels of TLR4,My D88,IκB and p-NFκB in liver and epididymis adipose tissue were analyzed by WB.Learning and immunohistochemistry to observe liver steatosis and inflammatory infiltration of epididymal adipose tissue.Results:(1)After 12 weeks of high-fat feeding,compared with the normal group,the model group had significantly higher body weight(p<0.01),fasting blood glucose(p<0.05),and serum insulin level(p<0.01).TC was significantly elevated(p<0.05),LDL-c level was significantly increased(p<0.05),impaired glucose tolerance,TG level in liver tissue was significantly increased(p<0.05),and a large number of lipid droplets appeared.After four weeks of administration,compared with the model group,the fasting blood glucose of the Jiaotai Pill group and the Coptidis group was significantly decreased(p<0.01),the serum insulin level was significantly decreased(p<0.01),and the LDL-c level was significantly decreased(p<0.05).The impaired glucose tolerancewas improved,and the TG level in liver tissue was significantly decreased(p<0.01).HE staining and oil red staining showed a marked decrease in lipid droplets and a marked improvement in steatosis.The above results indicate that Jiaotai Pill and Coptidis have the effect of improving the abnormality of glycolipid metabolism induced by high fat.(2)Compared with the normal group,the serum endotoxin level in the model group was significantly increased(p<0.01),the MCP-1 level in the epididymal adipose tissue was increased(p<0.01),and the TNF-α in the epididymal adipose tissue and liver tissue was significantly increased.(p<0.05),IL-6 increased significantly(p<0.01).Morphology and F4/80 immunohistochemistry showed that inflammatory cells infiltrated in epididymal adipose tissue,and a large number of corolla-like structures and epididymal adipose tissue appeared in the interstitium.The number of F4/80+ cells in liver tissue was significantly increased(p<0.01).After four weeks of administration,compared with the model group,the Jiaotai Pill group,the Coptidis group and the Bifidobacterium group could significantly reduce the serum endotoxin level(p<0.01),reduce the epididymal adipose tissue and liver tissue inflammatory factor levels and inflammatory cells.Infiltration.The above results indicate that Jiaotai Pill,Coptidis and Bifidobacterium have the effects of lowering serum endotoxin levels and improving chronic low-level systemic inflammation in liver and epididymal adipose tissue.(3)Western blot results showed that the expression levels of TLR4,My D88 and p-p65 protein in the liver and epididymis adipose tissue of the model group were significantly higher than those in the normal group(p<0.05),but the Jiaotai Pill group,the Coptidis group and the model.The expression levels of TLR4,My D88,and p-p65 protein were significantly lower than those of the group(p<0.05).The above results indicate that Jiaotai Pill and Coptidis have the effect of reducing the expression levels of TLR4,My D88 and p-NFκB on the TLR4/NF-κB signaling pathway.Conclusion:Jiaotai Pill and Coptidis improved chronic low-grade inflammation in mice induced by high fat diet and improved glucose and lipid metabolism. |
