Objective:To explore the wake-promoting effects of Deep brain stimulation(DBS)in traumatic brain injury induce-coma rats as well as the function of Orexin-A/OX1 R in the above effects.Method:54 SD rats were randomly divided into three group: Blank control group,control group(TBI)and test group(TBI+DBS).Traumatic brain injury induced comatose rats was established by free fall drop model.Consciousness level was evaluated via Sensory and motor movement assessment.Additionally,western blot and immunohistochemistry technique were used to detect the expression of Orexins receptor type 1(OX1R)level in the prefrontal cortex(PFC).Result:16 of 18 rats exhibited righting reflex in test group,and 7 of 18 rats had the same responses in control groups.Orexin-A and OX1 R levels in test groups were higher than control groups in PFC(p<0.05);Orexin-A and OX1 R levels in blank control groups were higher than control groups in PFC(p<0.05).Conclusion:DBS could promote recovery of consciousness in TBI induced comatose rats,and the potential mechanism may be due to upregulation of OX1 R in the PFC.
