Objective:To observe the protective effect of Farrerol on cisplatin-induced Mouse tubular epithelial cells injury Methods:(1)CCK-8 method was used to detect the cellular activity of renal tubular epithelial cells in mice treated with different concentrations of cisplatin and farrerol.Determining the appropriate concentration range for farrerol application and its protective effect on cells and establish the models of renal tubular epithelial cell injury.(2)The active oxygen scavenging activity of farrerol was determined using the oxidant-sensitive fluorescent probe DCFH-DA.(3)Detection of farrerol and p-NF-κB,NLRP3,p-p53,Bcl2,Bax and cleaved caspase-3 by Western-blot method,identifying farrerol’s links to cellular inflammation and apoptotic pathways(4)Hoechst 33342 / PI staining and flow cytometry analysis were used to detect the relationship between farrerol and apoptosis.(5)The statistical method used one-way analysis of variance,and the significance level was set to 0.05.Results:(1)The results of CCK-8 showed that farrerol concentrations at 5 μM,10 μM,and 20 μM had no effect on MTEC’s activity(P> 0.05),and Cisplatin has an effect on MTEC activity(P <0.05),And with the increase of cisplatin concentration,the stronger the cytotoxic effect has been showed,IC50 is 20μM.(2)Farrerol can reduce the cytotoxic effect caused by cisplatin,And as the concentration of farrerol increased,the effect of reducing the toxicity of cisplatin on cells was stronger,the effect is prominent when the farrerol concentration is 20 μM.(3)Farrerol can reduce the ROS produced by cisplatin And with the increase of farrerol’s concentration,the less ROS is produced.(4)At 5μM,10μM,and 20μM concentrations of farrerol the expression of p-NF-κB,and NLRP3,was reduced,As the concentration increases,the inhibition of protein expression becomes stronger.(5)Farrerol reduces the expression of P-P53,Bax,and caspase-3 at concentrations of 5 μM,10 μM,and 20 μM,as the concentration increased,the inhibitory effect on the expression become stronger(P<0.05);farrerol can enhance the expression of Bcl-2 protein at the concentration of 5μM,10μM,and 20μM,and as the concentration of rhododendron increases,the promotion of the protein expression becomes stronger(P<0.05).(6)Farrerol can reduce the number of apoptotic cells at concentrations of 5 μM,10 μM,and 20 μM.As the concentration increases,the number of apoptotic cells gradually decreases.Conclusion:(1)Farrerol could reduce the cytotoxic effects caused by cisplatin.(2)Farrerol can effectively reduce ROS produced by cisplatin and play a protective role on MTEC.(3)Farrerol could reduced the protein expression of p-NF-κB and NLRP3 to inhibit cisplatin-induced inflammation.(4)Farrerol could attenuate the expression of p-p53,Bax,and cleaved caspase-3 inhibited apoptosis...
