Experiment 1: Effect of Astragalus on proliferation and adhesion of human keratinocytes Objective:Through the experimental study on the effect of Astragalus membranaceus on the proliferation of human keratinocytes and the expression of β-catenin,the mechanism of Astragalus membranaceus in the treatment of psoriasis was preliminarily explored.Method:The effective concentration range of Astragalus in inhibiting the proliferation of HaCaT cells was detected by cell count method;the effect of Astragalus of different concentrations on the proliferation of HaCaT cells was detected by Cell Counting Kit-8;the expression ofβ-catenin mRNA was detected by real-time quantitative PCR;the expression of β-catenin was detected by Western blot.Result:(1)HaCaT cells were incubated with Astragalus injection of 0,0.01,0.1,1,10 and100mg/ml for 24 hours,the results showed that compared with the control group,Astragalus had obvious inhibitory effect on the proliferation of HaCaT cells after 1mg/ml;(2)According to the above results,the concentration range is about 1-10mg/ml,the experimental group was set as the Astragalus treatment group of 0.5,1,2,4,8,16 mg/ml after 24 and 48 hours.The CCK-8 kit test results showed that compared with the control group,the inhibition effect on the proliferation of HaCaT cells was significant when cells were treated with 1mg/ml Astragalus injection after 24 and 48 hours,and the difference was statistically significant;(3)The results of real-time quantitative PCR showed that compared with the controlgroup,the expression of β-catenin mRNA in HaCaT cells treated with 1 mg/ml Astragalus injection for 24 and 48 hours decreased significantly at 48 hours;(4)Western blot showed that compared with the control group,the expression ofβ-catenin in HaCaT cells treated with 1 mg/ml Astragalus injection for 48 hours decreased significantly.Conclusion:The mechanism of Astragalus in the treatment of psoriasis may be related to inhibiting the proliferation of human keratinocytes and the expression of β-catenin.Experiment 2: Effect of Bozhi Glycopeptides on the proliferation and adhesion of human keratinocytesObjective:Through the experimental study on the effect of Bozhi Glycopeptides on the proliferation of human keratinocytes,the mechanism of Bozhi Glycopeptides in the treatment of psoriasis was preliminarily explored,which provided experimental basis for the treatment of psoriasis with traditional Chinese medicine.Method:The CCK-8 kit was used to initially detect the effective concentration range of Bozhi Glycopeptides in inhibiting the proliferation of HaCaT cells;CCK-8 kit was used to detect the effect of different concentrations of Bozhi Glycopeptides injection on the proliferation of HaCaT cells;real-time quantitative PCR was used to detect the expression of β-catenin mRNA;Western blot was used to detect the expression of β-catenin.Result:(1)The results of CCK-8 kit test showed that compared with the control group,0.011719,0.023438,0.046875,0.09375,0.1875,0.375,0.75 and 1.5mg/ml Bozhi Glycopeptides for 24 hours had a tatistically significant inhibitory effect on the proliferation of HaCaT cells,and the effect of 0.023438mg/ml is the most significant;0.011719,0.023438,0.046875 and 0.09375mg/ml Bozhi Glycopeptides after 48 hours had a tatistically significantinhibitory effect on the proliferation of HaCaT cells;(2)According to the preliminary test results of CCK-8 kit,the concentration gradient was set as 0.625,1.25,2.5,5,10,20,40 and 80 ug/ml.The results showed that compared with the control group,different concentration of Bozhi Glycopeptides injection had inhibitory effect on the proliferation of HaCaT cells at 24 and 48 hours,the difference was statistically significant,and the inhibitory effect was the most significant after 20 ug/ml treatment for 48 hours;(3)The results of real-time quantitative PCR showed that compared with the control group,the expression of β-catenin mRNA in HaCaT cells treated with 10 and 20 ug/ml Bozhi Glycopeptides injection for 24 and 48 hours decreased significantly,and the effect was the most significant at 48 hours;(4)Western blot showed that the expression of β-catenin in HaCaT cells treated with 10ug/ml Bozhi Glycopeptides injection for 24 hours decreased significantly compared with the control group.Conclusion:The mechanism of Bozhi Glycopeptides in the treatment of psoriasis may be related to inhibiting the proliferation of human keratinocytes and the expression of β-catenin.
