In China,lung cancer has become one of the malignant tumors with the highest incidence and mortality.Because lung cancer has a very complicated mechanism of occurrence and development,the molecular mechanism of its exact pathogenesis is still in the exploration stage.TMEM16A(Transmembrane protein 16A)is the molecular basis of calcium-activated chloride channels Ca CCs(Calcium-activated Chloride Channels),widely distributed in various tissues of mammals.Recent studies have found that dysregulated expression of TMEM16 A is related to the occurrence and development of cancer.In this study,the bioinformatics series of online software was used to systematically analyze the correlation between TMEM16 A,miR-381,and TMEM16 A and cancer.Real-time quantitative PCR,Western blot,cellular immunofluorescence,CCK,scratches,epithelial-mesenchymal(EMT)and Transwell methods were used to analyze the expression and localization of TMEM16 A in non-small cell lung cancer cell lines,and systematically explored miR-381 and TMEM16 A expression correlation and regulatory relationship,analyzed the miR-381 targeted regulation of TMEM16 A expression on the proliferation,migration and invasion of non-small cell lung cancer.Bioinformatics analysis found that TMEM16 A was the predicted target gene of miR-381,and there was a miR-381 binding site in the 3’-UTR of TMEM16 A.Therefore,TMEM16 A may be a direct target of miR-381,and TCGA analysis found that TMEM16 A was It has an important influence in the occurrence and development of cancer.Western Blot and real-time quantitative PCR results showed that the expression of TMEM16 A in non-small cell lung cancer cell lines was higher than that of normal lung epithelial cells,and cell immunofluorescence experiments showed that TMEM16 A was mainly expressed in the cell membrane and cytoplasm.But the difference between lung adenocarcinoma and lung squamous cell carcinoma is not obvious.Fluorescence real-time quantitative PCR detection found that the expression level of miR-381 in non-small cell lung cancer cell lines was significantly lower than that of normal lung epithelial cell lines;the results of fluorescence real-time quantitative PCR and Western Blot showed that miR-381 and TMEM16 A werenegatively regulated Relationship,increasing the expression of miR-381 in non-small cell lung cancer cell lines can significantly reduce the expression of TMEM16 A in m RNA and protein levels in non-small cell lung cancer cell lines.The experimental results of CCK,scratch,EMT,Western Blot and Transwell show that miR-381 can target negatively regulate the expression of TMEM16 A and effectively inhibit the progress of non-small cell lung cancer in proliferation,migration and invasion.In summary,this study provides new clues for the development of targeted drugs and targeted therapies for non-small cell lung cancer.
