Objective The purpose of this study was to observe the liver,skeletal muscle and ovarian insulin receptors(InsR),insulin receptor substrate-1(IRS-1),and phosphorylated insulin receptor substrate-1(P-IRS-1)in hyperinsulinemia mice fed by high-fat and high-sugar.The expression levels of InsR,IRS-1 and P-IRS-1 in the liver and skeletal muscle of hyperinsulinemia mice fed with high-fat and high-sugar glucose gradually decreased with the progress of insulin resistance.The distribution of insulin receptors gradually decreased.Methods 60 healthy4-week-old female C57 BL / 6J mice were randomly divided into a ordinary feed group(NF group)and a high-fat and high-sugar group(HF group).There are 6 groups of 10 mice each.The first three groups are all NF groups,and the last three groups are all HF groups.Among them,the ordinary feed feeding group and the high-fat and high-sugar group correspond to three groups of the NF group and the HF group according to the length of feeding were named as the feeding 0 week group,the feeding 6 week group,and the feeding 12 week group.At 0,6,and 12 weeks,the weight of each group of mice was measured,and blood was collected from the tail vein to measure fasting blood glucose.Enzyme linked immunosorbent assay(ELISA)was used to determine the total cholesterol,triglyceride,and Insulin index and calculation of insulin resistance index;qPCR was used to measure the expression levels of InsR and IRS-1 mRNA in liver,skeletal muscle and ovary of each group.Western Blot was used to compare liver and skeletal muscle of each group And ovarian InsR,IRs-1,P-IRs-1level gray value,so as to explore the changes in the expression levels of InsR,IRS-1,P-IRs-1in each tissue in each period.Results 1.The body weight,fasting blood glucose,total cholesterol,triglyceride,insulin and insulin resistance index of mice in the HF group fed for 6 and 12 weeks were significantly larger than those of the mice in the NF group fed for 6 and 12 weeks.That is,after 6 weeks of feeding,the mice in the HF group had an insulin resistance state.After 12 weeks of feeding,the IR stat was more stable and the model was successfully established.2.The expression of InsR and IRS-1 mRNA in liver and skeletal muscle of HF group mice at 6 and 12 weeks was significantly lower than the mRNA expression of NF group mice at 6 and 12 weeks.The mRNA expression of InsR and IRS-1 in ovarian tissue of HF group significantly higher than the NF group at 6 and 12 weeks.The differences in liver,skeletal muscle,and ovary of the two groups of mice became more and more obvious with time.3.The gray levels of InsR,IRs-1,and P-IRs-1 in liver and skeletal muscle mice in the HF group at 6 and 12 weeks were significantly lower than those in the NF group at 6 and 12 weeks.The gray level of IRs-1 was higher than that of NF group at 6 and 12 weeks.Conclusion The expression levels of InsR,IRS-1 and P-IRS-1 in the liver and skeletal muscle of hyperinsulinemia mice fed with high-fat and high-sugar glucose gradually decreased with the progress of insulin resistance The distribution of insulin receptors gradually decreased.The expression levels of InsR,IRS-1 and P-IRS-1 in ovary tissues gradually increased with the progress of insulin resistance and the distribution of insulin receptors gradually increased.
