Objective:To investigate the effect of miR200 c target gene PCDH8 on the proliferation,migration and angiogenesis of esophageal squamous cell carcinoma(ESCC).Methods:The candidate mir-200 c target gene PCDH8 was selected by using the miRNA target gene prediction software including of TargetScan and PicTar.Luciferase reporter gene assay verified whether PCDH8 was the target gene of miR-200 c.Immunohistochemistry was used to detect the expression of PCDH8 in esophageal squamous cell carcinoma and analyze its relationship with clinicopathological parameters.PCDH8 and miR-200 c expression levels of KYSE-150 cell line and normal esophageal squamous epithelial HET-1A cell line were detected by qPCR and Western blotting.PCDH8 gene overexpression vector was transfected into ESCC cell line KYSE-150 to observe the changes of proliferation,migration,invasion ability and angiogenesis of KYSE-150 cells.Results:(1)Luciferase reporter gene assay confirmed that PCDH8 was the target gene of miRNA-200c;(2)Compared with the control group,the expression of PCDH8 in esophageal squamous cell carcinoma was decreased(P<0.05),and the expression of PCDH8 was correlated with tumor size(P value 0.011,<0.05),histological grade(P value 0.002,<0.01)and lymph node metastasis(P value 0.017,<0.05);(3)Compared with HET-1A cells,the expression of miR-200 c in KYSE-150 cells was significantly increased(P<0.05),while the expression of PCDH8 was decreased(P<0.05);(4)CCK8 assay showed that the proliferation of KYSE-150 cells overexpressing PCDH8 was inhibited;(5)Transwell assay showed that PCDH8 overexpression significantly reduced the invasion ability of KYSE-150 cells;(6)Quantitative ELISA showed that VEGFA expression was down-regulated in PCDH8 overexpressed KYSE-150 cells;(7)The angiogenesis experiment showed that the overexpression of PCDH8 decreased the angiogenesis of kyse-150 cells.Conclusion:(1)PCDH8 is the target gene of miR-200 c,which inhibits the expression of PCDH8 in esophageal squamous cell carcinoma;(2)In esophageal squamous cell carcinoma,miR-200 c expression was up-regulated while PCDH8 expression was down-regulated,suggesting that miR-200 c and PCDH8 were involved in the occurrence and development of esophageal squamous cell carcinoma;(3)KYSE-150 cell line transfected with PCDH8 was found that its proliferation,invasion and angiogenesis were decreased,while its apoptosis was increased,suggesting that PCDH8 may play an oncogene inhibitory role in esophageal cancer.
