Objective: Immunohistochemistry and RT-PCR were used to detect the expression levels of hypoxia-induced factor-2α(HIF-2α)and vascular endothelial growth factor(VEGF)in the cancer tissues and adjacent tissues of Tibetan and han CRC patients in Qinghai To analyze the differences of expression of HIF-2α and VEGF in cancer tissues and paracancer tissues of han nationality and Tibetan CRC patients.To investigate the relationship between HIF-2α and VEGF expression,clinicopathology and prognosis in CRC tissues of han nationality and Tibetan nationality.Methods: collected in October 2017-November 2019 in qinghai university hospital after colonoscopy examination and pathological examination diagnosis of colorectal cancer and meet the criteria for the 70 cases of Tibetan and han ethnic group,80 cases as the research object,using immunohistochemical method of HIF-2α and VEGF proteins in CRC tissue adjacent to carcinoma tissues and its corresponding expression level of testing,application method of polymerase chain reaction(RT-PCR)detection in the CRC tissue adjacent to carcinoma tissue and its corresponding HIF-2α and VEGF m RNA transcription level.Results: 1.There was no statistically significant difference in the expression of HIF-2α and VEGF proteins in the paracancer tissues of Tibetan and han patients(P>0.05).2.There was no statistical difference in the expression of HIF-2α and VEGF proteins in colorectal cancer tissues of Tibetan and han nationality(P>0.05).3.There was no statistically significant difference in HIF-2α and VEGF m RNA transcription levels in colorectal cancer tissues of Tibetan and han patients(P>0.05).4.The expression of HIF-2αand VEGF proteins in colorectal cancer tissues was significantly higher than that in para-carcinoma tissues(P<0.05).5.The m RNA transcription levels of HIF-2α and VEGF in colorectal cancer tissues were much higher than that in normal tissues(P<0.05).6.The survival status of patients was correlated with tumor stage(TNM)(P<0.05),but not with age,gender,pathological differentiation and tumor size(P > 0.05).7.HIF-2αand VEGF protein expression and m RNA transcription level in colorectal cancer patients were correlated with TNM stage(P<0.05).Conclusion: 1.There was no significant difference in HIF-2 α and VEGF expression between Tibetan and han CRC patients.2 The expression of HIF-2αand VEGF in colorectal cancer tissues in Qinghai was significantly higher than that in paracarcinoma tissues,suggesting that HIF-2α and VEGF may play a crucial role in tumor development and metastasis.3.TNM staging of CRC tumors directly affects the survival status and prognosis of patients.
