| Objective 1.The mouse models of different doses of acute radioactive heart injury were established in clinical simulation,and the scientific rationality of the mouse models was evaluated to provide a feasible animal model for the next experiment.?2.To study whether Gastrodin has protective effect on mice with radioactive heart injury.Methods 1.25 C57 BL / 6 mice were randomly divided into control group,6 Gy,12 Gy,18 Gy,30 Gy irradiation group in addition to the control groups of mice by using medical linear accelerator radiation doses of radioactive heart damage in mice model was established.At 30 days after irradiation,with ELISA method to detect the contents of serum IL-6 in mice,Western blot method to detect myocardial tissue caspase 3 levels,colorimetry to detect superoxide dismutase(SOD),malondialdehyde(MDA)level,take part of the heart tissue and HE staining.2.50 C57BL/6 mice were randomly divided into control group,simple irradiation group,irradiation + Gastrodin low,medium and high dose intervention group(50,100,150 mg/kg).Mice in each experimental group were modeled using a radiotherapy accelerator(radiation dose 18 Gy).Mice in each drug intervention group were intraperitoneally injected with Gastrodin solution once a day from 3 days before irradiation.At 30 days after irradiation,the levels of the above related indicators in mice were detected by the same detection method as that in 1.Results 1.Compared with the control group,the levels of il-6 caspase-3 MDA and SOD in mice of each exposure group were significantly increased,with statistically significant differences(P<0.05).Comparison between multiple groups of each exposure group: the difference level of the above indicators was statistically significant in the 18 Gy group and the 6Gy 12 Gy group(P<0.05).Comparison between the 18 Gy group and the 30 Gy group showed no statistically significant difference in the above indicators(P>,0.05).HE staining: with the increase of irradiation dose,the degree of myocardial cell damage of mice in each irradiation group increased gradually.2.Compared with the control group,the expression level of il-6 caspase-3 MDA was significantly increased and SOD was significantly decreased in mice in the group exposed to radiation alone,with statistically significant differences(P<0.05).Compared with the simple exposure group,the expression of il-6 caspase-3 and MDA in the medium and high dose drug intervention group was significantly decreased,while the expression of SOD was significantly increased,with statistically significant difference(P<0.05).Compared with the single irradiation group,the expression levels of il-6,caspase-3,SOD and MDA in the low-dose drug intervention group were not statistically significant(P >,0.05).HE staining: Gastrodin intervention could reduce the degree of pathological damage of myocardial tissue in mice,especially in the medium and high dose drug intervention group.Conclusion 1.Animal models of radioactive heart injury in mice were successfully established at different doses.Among the four doses of 6,12,18,30 Gy,we believe that single dose of 18 Gy is suitable for the establishment of animal models of radioactive heart injury in mice;Inflammation,apoptosis of cardiomyocytes,and oxidative stress play important roles in mouse RIHD.2.Gastrodin can play a protective role in radioactive heart injury of mouse cardiomyocytes by down-regulating the degree of oxidative stress response of cardiomyocytes,reducing apoptosis and anti-inflammation. |
