| Objectives:To observe the regulatory effect of Echinacea on the proteins related to IRE1α mediated the promotion of apoptosis signal pathway in rotenone induced PD rats,and its effect onα-synuclein and TH,which are closely related to Parkinson’s disease,and to explore the related mechanism of the protective effect of Echinacea on neurogenesis.Method:In the modeling stage,PD rats were established by subcutaneous injection of rotenone sunflower oil emulsion at the back of the neck.The rats were randomly divided into model group and treatment group,and the solvent control group(subcutaneous injection of sunflower oil emulsion on the back of neck)and normal group(not treated)were added.During the drug intervention stage,the treatment group was given a gavage of Echinacea solution,and the rest group was given normal saline the same volume for 14 days.Observing the behavioral performance of rats;the expression of th,ASK1,caspase-12 in immunohistochemical test;the expression of the α-synuclein,IRE1α-ask1-jnk in immunofluorescence test;the expression of the IRE1α-ask1-jnk signaling path that related apoptin in Western blot detection.Results:1.The results of stride test showed that the stride of model group and treatment group was shorter than that of normal group and solvent control group(P < 0.05);the stride length of treatment group was longer than that of model group,but there was no statistical significance(P > 0.05).2.The results of suspension test showed that the suspension time of model group and treatment group was shorter than that of normal group and solvent control group(P < 0.05);compared with model group,the suspension time of treatment group was longer(P < 0.05).3.The results of immunofluorescence showed that compared with the normal group and the solvent control group,the expression of α-synuclein in the substantia nigra and hippocampus increased in the model group and the treatment group(P < 0.05);compared with the model group,the expression of α-synuclein in the substantia nigra and hippocampus decreased in the treatment group(P < 0.05).4.The results of immunohistochemistry showed that compared with the normal group and the solvent control group,the TH positive cells in the substantia nigra of the model group and the treatment group decreased(P < 0.05),the expression of ASK1 and caspase-12 in the model group and the treatment group increased(P < 0.05);compared with the model group,the TH positive cells in the substantia nigra of the treatment group increased(P < 0.05),and the expression of ASK1 and caspase-12 in the treatment group was decreased(P < 0.05).5.Western blot showed that the expression of p-ire1α / IRE1α,GRP78,ASK1,Caspase-12,p-JNK / JNK in the model group was increased which compared with the normal group and the solvent control group(P <0.05);and the treatment group increased compared with the normal group and the solvent control group(P < 0.05);compared with the model group,the expression of p-ire1 α / IRE1 α,GRP78,ASK1,caspase-12,p-JNK / JNK in the treatment group was decreased(P < 0.05).Conclusion:1.To some extent,Echinacea can improve the behavior of PD rats induced by rotenone,increase the th content of PD rats,inhibit the accumulation of α-synuclein in the midbrain,and play a neuroprotective role.2.The neuroprotective effect of Echinacea on PD model rats may be related to its inhibition of IRE1 which mediated the promotion of apoptosis signal pathway that related protein expression. |
