Purpose:In order to find the noninvasive biomarkers with high specificity and sensitivity for early diagnosis of cervical cancer,the early diagnostic value and clinical significance of mi R-141、mi R-192、mi R-99a、mi R-153、mi R-24、mi R-25、mi R-30a、mi R-335、mi R-196b、mi R-29 a and mi R-1290 in the serum of the cervical cancer were explored.Method:We selected fasting peripheral venous blood samples of the patients who were initially diagnosed cervical cancer(CC)and cervical intraepithelial neoplasia(CIN)before treatment in Shaanxi cancer Hospital from October 2018 to September 2019,including 62 cases of cervical cancer and 22 cases of cervical intraepithelial neoplasia,all of which were confirmed by pathological examination.Cervical cancer patients were phased according to the latest criteria of International Federation of Gynecology and Obstetrics(FIGO).At the same time,72 healthy volunteers were taken as the normal control group.Real-time fluorescence quantitative polymerase chain reaction(q RT-PCR)was used to test the relative expression of each mi RNA in serum of normal control group,and patients with cervical cancer or CIN respectively.The data were analyzed by SPSS 25.0 statistical software.The mean±standard deviation(M±SD)was used to express measurement data.T-test was used to analyse inter-group correlation.The relationship between abnormal serum mi RNA and cervical cancer was analyzed by binary Logical regression.The expression of mi RNA was compared with the clinical parameters of cervical cancer by Mann-Whitney test.The diagnostic value of each mi RNA in serum of cervical cancer was evaluated by drawing the receiver operating characteristic(ROC)curve.P-value less than 0.05 was considered to be significantly different between groups.P-value less than 0.01 or 0.001 were considered to be remarkably significant between groups.Results:1.The expression of mi R-141,mi R-192,mi R-99 a,mi R-153,mi R-24,mi R-25,mi R-30 a,mi R-335,mi R-196 b and mi R-29 a significantly decreased in the serum of the patient with cervical cancer compared with the normal control group(P<0.05).All differences are statistically significant except for mi R-192、mi R-99a、mi R-24 and mi R-29 a in CIN.However,the expression of mi R-1290 significantly increased in the serum of the patients with cervical cancer and CIN(P<0.05).Both differences are statistically significant.2.As the result of the binary Logical regression showed that the down-regulation of mi R-141,mi R-192,mi R-99 a,mi R-153,mi R-24,mi R-25,mi R-30 a,mi R-196 b,mi R-29 a and mi R-335 were the independent risk factors in the serum of the cervical cancer(P<0.05).However,the up-regulation of mi RNA-1290 in the serum was the independent risk factor of the cervical cancer(P<0.05).3.We analyzed the relationship between the expression of those 11 mi RNAs and the clinical parameter respectively,including age,FIGO stage,tumor size,the degree of the differentiation,lymphatic metastasis and intravascular infiltration,and found that the low expression of mi R-99 a in serum of the patients with cervical cancer was negatively related to tumor size(P=0.012),as well as lymphatic metastasis(P=0.017),but no correlations were detected in another clinical pathological parameters(P >0.05).The low expression of mi R-196 b in serum of the patients with cervical cancer was negatively correlated with the degree of tumor differentiation(P=0.036),but no correlations were detected in another clinical pathological parameters(P>0.05).The high expression of mi R-1290 in the serum was positively correlated with FIGO stage(P=0.021)and tumor size(P=0.010),but no correlations were detected in other clinical pathological parameters(P>0.05).4.The diagnostic value of mi R-141,mi R-192,mi R-99 a,mi R-153,mi R-24,mi R-25,mi R-30 a,mi R-335,mi R-196 b,mi R-29 a and mi R-1290 were evaluated by the ROC curve.We found that the diagnostic specificity and sensitivity of mi R-99 a were 84.7%and 53.2% respectively.The diagnostic specificity of mi R-1290(86.1%)was the highest,but its sensitivity(35.5%)was a little low.The Youden’s index(0.623)and the area under the ROC curve(0.852)of mi R-196 b were the largest among them and the specificity and sensitivity were 83.3% and 79.0% respectively.In one word,mi R-196 b can be considered as the potential noninvasive biomarker in the diagnosis of the cervical cancer.Conclusion:1.The negative correlation was found between the expression of mi R-141,mi R-192,mi R-99 a,mi R-153,mi R-24,mi R-25,mi R-30 a,mi R-335,mi R-196 b and mi R-29 a and the development and progression of the cervical cancer.However,the positive correlation was found between the expression of mi R-1290 and the development and progression of the cervical cancer.2.The mi R-196 b is expected to be a noninvasive biomarker for the early diagnosis of cervical cancer. |
