The Network Pharmacology Research Of "Fuzheng-Quxie" Mechanism Of Shenmai Injection Against Lung Cancer

Objective: Aimed at the anti-lung cancer mechanism of "Fuzheng-Quxie" of Shenmai injection,investigation has been carried out by using the Network pharmacology.Methods:In the CNKI database literature search,the main active ingredients of Shenmai Injection were excavated,and its three-dimensional structure was obtained from the ChemalBook database and stored as mol2 files.First,the potential target genes of the main components of Shenmai Injection was predicted by using the SwissTarget Prediction data platform.And then,the genes highly related to lung cancer in the GeneCards and DisGeNET gene banks were screened.Second,the "Shenmai Gene-Lung Cancer Gene" interaction network was constructed using the STRING database,then the direct anti-cancer targets and network center targets of Shenmai Injection can been screened.Finally,the function analysis of Gene Ontoology of direct anti-cancer targets and network center targets was conducted using CueGO software.The key target KEGG pathway was analyzed through the DAVID V6.8 platform.Results:Shenmai Injection regulates 25 lung cancer target genes such as AKT1,CASP3,MAPK1,MMP9,RB1,STAT3,VEGFA,SRC,NOS3,etc.The anti-Lung Neoplasms pharmacological effects are realized by regulating 5 kinds of 22 pathways,(1)VEGF and HIF-1 contral Cancer cell growth;(2)MAPK,Ras,PI 3K-Akt,ErbB,mTOR,FoxO,etc pathways inhibiting tumor proliferation;(3)Adherens junction,Gap junction,Focal adhesion inhibiting tumor infiltration metastasis;(4)Tumor necrosis factor,NK natural killer cells,T cell receptors,improved B cell receptor regulating tumor immune function,(5)other tumor pathways related to tumor metabolism,etc;to realize direct anti-cancer,regulating tumor immune and other pharmacological effects.Conclusion: Shenmai injection can inhibit the proliferation,development,infiltration and metastasis of lung cancer through "multiple components,multiple targets,and multiple pathways",inhibit tumor angiogenesis,achieve the "Qu xie" effect,and achieve the "positive" effect by regulating the body’s tumor immunity.
