| Objective:Hepatocellular carcinoma(HCC)is a common malignant tumor of digestive sys-tem.Hepatitis B virus(hepatitis B virus,HBV)infection is an important risk factor for HCC,but the exact mechanism of HCC is not clear.The relationship between chronic inflammation and tumor has always been one of the hotspots in the field of tumor biology.Interleukin-6(Interleukin-6,IL-6),as an important inflammatory factor,par-ticipates in a variety of inflammatory and immune response processes,plays an im-portant role in tumor microenvironment,and is closely related to the occurrence and development of tumor.In this study,the clinical information of patients with liver cancer and liver cancer samples were studied to verify the expression of IL-6 in liver cancer and para-cancerous tissues,and to explore the differences of IL-6 expression in liver cancer and para-cancerous tissues and the effect of hepatitis B positive on IL-6expression.Methods:The target genes of mi RNA were predicted by IPA analysis to explore the close relationship between the potential related pathway and hepatitis B positive liver cancer.Through the analysis of liver cancer sample data in TCGA database,the expression of IL-6 in liver cancer was predicted.Through retrospective analysis of previous cases of hepatocellular carcinoma,a total of 37 cases were selected to meet the study criteria.The clinicopathological data of the patients were collected,and the clinical data of the patients were counted and analyzed.QRT-PCR and immunohistochemical techniques were used to analyze the expression of IL-6 in tumor and para-cancerous tissues,and to explore the relationship between the expression of IL-6 in para-cancerous tissues and a series of related clinical information of patients.The m RNA and protein expres-sion levels of IL-6 in HBV positive group and HBV negative group were compared.Results:In IPA,the IL-6 signal pathway was significantly enriched in the signal pathway of|Z-score|≥0.According to the data analysis of TCGA database,the m RNA expression of IL-6 in para-cancerous tissues was higher than that in HCC tissues,but there was no statistical significance(p>0.05).In the q RT-PCR experiment,the expression of IL-6m RNA in HCC tissues was significantly higher than that in para-cancerous tissues,(p<0.05).The expression level of IL-6 m RNA in para-cancerous tissues was correlated with the size of tumor((p<0.05,r>0)),but not with age,sex,number of tumors,presence of portal vein tumor thrombus,liver cirrhosis,CNLC stage and serum AFP(p>0.05).There was no correlation between the expression of IL-6 m RNA and the positive expression of HBV in HCC(p>0.05).Compared with the HBV-DNA negative group,the expression of IL-6 m RNA in the HBV-DNA positive group was down-regulated(p<0.05).Com-pared with HBV positive liver function normal group and HBV negative liver function normal group,the relative differential expression of IL-6m RNA in HBV positive liver function abnormal group was significantly higher than that in HBV positive liver function normal group and HBV negative liver function group(p<0.05).In the immunohisto-chemical experiment,the positive expression rate of IL-6 protein in para-cancerous tis-sues was significantly higher than that in HCC tissues(~2=35.764,P<0.01).The ex-pression of IL-6 protein in HCC was not correlated with the positive expression of HBV,HBV-DNA and the level of liver transaminase(p>0.05).Conclusion:IL-6 signal pathway may be closely related to hepatitis B infection and the occur-rence and development of liver cancer.Compared with HCC tissues,the expression of IL-6 m RNA and protein in para-cancerous tissues were up-regulated,and the expression of IL-6 m RNA in para-cancerous tissues was related to tumor size.The expression of IL-6 m RNA in HCC tissues was negatively correlated with the replication level of HBV-DNA,and the level of Hepatic transaminase was positively correlated with the relative amount of IL-6 m RNA in liver cancer.These results suggest that IL-6 may mediate the occurrence and development of HCC through a potential pathway. |
