| Objective: Myofascial pain syndrome(MPS)is caused by trigger points of myofascial fascia.It is manifested as local tenderness and involved pain in soft tissues and clinical symptoms related to sensory,motor,and autonomic nervous systems.With the development of society,the increase of working hours at desks and the acceleration of life rhythm,myofascial pain syndrome has become one of the common causes of non-articular skeletal muscle pain.In the clinic,the floating needle has achieved good results in treating myofascial pain syndrome,but its treatment mechanism is rarely reported.In this study,the observation of the floating needle intervention on tumor necrosis factor(TNF-α),5-hydroxytryptamine(5-HT),c Calcitonin gene related peptide(CGRP)expression and histomorphological changes,preliminary exploration of the mechanism of floating needle treatment of myofascial pain syndrome.Methods: Forty-eight healthy male SD rats were randomly divided into 2 groups,each group containing 24 rats.(1)NC(Normal Control Group)24 animals,6 animals were sacrificed after one week of adaptive breeding,biochemical indicators were tested,and the rest of the time we’re given stimulation.(2)MC(Myofascial pain Control Group)24 animals,on the first day of each week,a heavy object with a kinetic energy of 2.352 J was struck on the left medial femoral muscle of the rat,and the second day was subjected to a centrifugal exercise on a-16 ° electric treadmill.Ninety minutes,and then reared normally.(3)After the modeling was completed,6 rats were randomly selected from group MC for Fu’s Subcutaneous Needling treatment(Fu’s Subcutaneous Needling Group,FC),three times a week for two weeks.In the first stage,at the fifth week and the eighth week,6 rats in each of the two groups of A and B were randomly sacrificed.The peripheral blood TNF-α and 5-HT contents of the rats were measured.Observe the morphological changes;In the second stage,the left medial femoral muscle of the rats was taken 2 weeks after the Fu’s Subcutaneous Needling treatment,and the morphological changes were observed by HE stainingsections.The 5-HT content in the brain tissue and peripheral blood was measured,and the peripheral blood TNF-α content and CGRP content of damaged tissues.Results:(1)Stage 1: In the fifth week,compared with the NC group,MC group5-HT,TNF-α,and 5-HT content in the brain tissue of rats were increased(P> 0.05),but there was no statistical significance.At the eighth week,the modeling was completed.Compared with the NC group,MC group 5-HT and TNF-α levels in the blood of the rats were significantly increased(P <0.05),and the 5-HT content in the brain tissue was significantly increased(P <0.05).Statistical differences.The femoral muscle HE staining showed that:(1)the muscle cell distribution was disordered,edema,penetrating cell infiltration was obvious,the nucleus moved inward,and formed a circular contraction and nodule,and(2)longitudinal section,irregular muscle fiber thickness It is fusiform,showing cleavage and fibrous middle adhesion and swelling,and thinner muscle fibers appear thinner,atrophy,breakage and distortion.(2)The second stage: the treatment was completed in the tenth week,and the proportion of the MC group,the FC group’s 5-HT,TNF-α,and the damaged tissue CGRP content were significantly reduced(P <0.05),and the brain tissue 5-HT The content was significantly increased(P <0.05).Compared with the blank group,the levels of blood 5-HT,TNF-α,and CGRP in the damaged tissue of the treatment group were significantly increased(P<0.05),and the content of 5-HT in the brain tissue was significantly increased(P<0.05).Compared with the blank group,there was no significant change in the content of each factor in the treatment group;at different time periods,the 5-HT and TNF-α levels in the blood of model rats increased together,showing a normal linear correlation(P <0.05),with statistical differences;The medial femoral muscle HE staining showed that:(1)On the cross section,muscle cell edema and inflammatory infiltration were all improved.Circular nodules still partially existed,and most muscle cells have been repaired.(2)On the longitudinal section,the muscle fibers are arranged slightly regularly,the muscle fibers that are broken and distorted are reduced,the spindle-shaped aggregate structure is reduced,and the damaged muscle fiber tissue has been repaired.Conclusions: 1.Fu’s Subcutaneous Needling intervention reduces the levels of serotonin and TNF,serotonin,and calcitonin gene-related peptides in the blood of the rats with myofascial pain syndrome model,which is beneficial for reducing pain.This may be related to floating needle pine.2.Fu’s Subcutaneous Needling treatment can improve the pathological state of injured muscles and promote the recovery of muscletissues in rats with myofascial pain syndrome. |
