Objective To investigate the association between Apa I polymorphism of VDR gene and type2 diabetes mellitus(T2DM)in the Mongolian and Han nationalities of Hohhot,Inner Mongolia.Methods From October 2017 to may 2019,Mongolian and Han diabetes patients who were treated in the first hospital of Hohhot,Inner Mongolia Autonomous Region Hospital and Affiliated Hospital of Inner Mongolia Medical University were selected as the case group,while Mongolian and Han healthy people who had physical examination in the hospital at the same time were selected as the control group.Polymerase chain restriction fragment length polymorphism(PCR-RFLP)and agar gel electrophoresis were used to detect the genotype and alleles of VDR gene Apa I locus in two groups.The distribution difference between the two groups was compared.Two groups of Logisitic regression analysis were used to take age as a dependent variable,and age,gender,body mass index,blood pressure,genotype,nationality and C peptide as the dependent variable.In the population with T2DM,ethnic groups were divided into Mongolian and Han,and multiple logistic regression model was used to analyze the expression differences of three basic types of Mongolian and Han in order to explore the pathogenesis of T2DM.Results(1)The frequency distribution of Apa I allele of VDR gene in the study subjects was in accordance with Hardy Weinberg law,indicating that the samples were in accordance with the law of heredity;(2)there was a statistical difference between the case group and the control group in the distribution frequency of Apa I genotype and allele of VDR gene(χ~2=14.503,χ~2=5.011,P<0.05);(3)the results of binary logistic regression analysis showed that:high diastolic pressure(OR=1.454,OR95%CI:1.046,2.020),Mongolian(OR=4.713,or95%CI:1.494,14.867),genotype aa(OR=3.965,OR95%CI:1.565,10.048),genotype Aa(OR=2.976,OR95%CI:1.353,6.546)was the risk factor of T2DM in this study;(4)the results of multi classification logistic regression analysis showed that the risk of T2DM in Mongolian with genotype aa was 1.826 times higher than that in Han with genotype aa(P<0.05).Conclusion(1)There is a correlation between Apa I locus of VDR gene and T2DM in Hohhot,Inner Mongolia.;(2)In Hohhot,Inner Mongolia,Mongolian people with aa genotypes have a higher risk of developing T2DM than Han people with aa genotypes. |
