Role Of Long Non-coding RNA HOTAIR In The Proliferation And Apoptosis Of Pancreatic Cancer Cells | | Posted on:2021-04-25 | Degree:Master | Type:Thesis | | Country:China | Candidate:T Sun | Full Text:PDF | | GTID:2404330614464443 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Objective To investigate the effect of HOTAIR gene on the biological behavior of human pancreatic cancer cells,and to explore the effect and mechanism of HOTAIR on the proliferation and apoptosis of pancreatic cancer cells.Methods The pancreatic cancer cell line PANC-1 cells with overexpression of HOTAIR were obtained by over expression lentivirus transfected,the efficiency of lentivirus transfection was detected by the fluorescence microscopy and real-time PCR.The MTT test was used to detect the proliferation of pancreatic cancer cell line PANC-1 cells,and the TUNEL staining experiments was used to detect the apoptosis of pancreatic cancer cell line PANC-1 cells,and the expression of mi R-214 was detected by real-time quantitative PCR.Results The mRNA expression of HOTAIR in pancreatic cancer cell line PANC-1 cells were significantly increased after transfected by the over expression lentivirus,the results indicated that a stable-transmitted pancreatic cancer cell line PANC-1 cells model with HOTAIR overexpression was successfully established;MTT detection of cell proliferation showed that after over-expressing HOTAIR,the proliferation ability of the experimental group was significantly enhanced compared to the control group;TUNEL staining experiments showed that after over-expression of HOTAIR,the experimental group’s mortality drops significantly;Real-time PCR showed that after overexpression of HOTAIR,the expression of mi R-214 in the test group was significantly lower than that of the control group PANC-1 cells.Conclusion The up-regulation of HOTAIR gene expression in pancreatic cancer PANC-1cells can efficaciously promote the proliferation of pancreatic cancer cells and inhibit the apoptosis of pancreatic cancer cells.The mechanism may be related to the expression of mi R-214 in pancreatic cancer cells.The HOTAIR plays an important role in the development of pancreatic cancer and may be a breakthrough for clinical diagnosis and treating pancreaticcancer. | | Keywords/Search Tags: | Pancreatic cancer, LncRNA, HOTAIR, miRNA-214, Proliferation, Apoptosis | PDF Full Text Request | Related items |
