Objective To investigate the ratio of CD4+IL-17+Th17 and CD4+CD25+Foxp3+Treg in CD4+ T lymphocytes of asthmatic model rats by acupuncture,and explore the effect of acupuncture of "three points and five needles" on asthma from the perspective of immune regulation Regulation of Th17/Treg balance in rat peripheral blood.Methods Thirty SPF male SD rats were adaptively fed and divided into blank group,model group and acupuncture group according to the randomization principle,10 rats in each group were given corresponding intervention,and materials were taken after anesthesia.Take rat lung tissue sections,observe after HE staining;take rat alveolar lavage fluid to detect white blood cell count;take rat peripheral blood to extract monocytes,and Detection of CD4+IL-17+Th17 and CD4+CD25+Foxp3+Treg in CD4+T lymphocytes by flow cytometry;Western Blot method was used to detect Foxp3 and STAT3 protein expression in rat lung tissue.Results 1 The behavioral results showed that the rats in the blank group had no obvious abnormal changes,and the rats in the asthma model group could observe different degrees of shortness of breath,sneezing,irritability,scratching,cyanosis of the mouth and nose after challenge.After acupuncture intervention,the symptoms of shortness of breath,cyanosis of mouth and nose,and irritability in the acupuncture group were improved.2 The white blood cell count showed that compared with the blank group,the white blood cell count in the model group was significantly increased(P<0.05),and that in the acupuncture group rising trend(P<0.05);compared with the model group,the white blood cell count in the acupuncture group showed a downward trend(P<0.05).3 Lung tissue HE showed that compared with the normal group,inflammatory masses dominated by eosinophils were seen in the model group,and epithelial cells were deformed and shed in the bronchial mucosa,while the number of inflammatory cells in the acupuncture group was Significantly reduced,and the edema of bronchial mucosa was significantly reduced.4 Flow cytometry detection of Th17/Treg ratio showed that comparison with blank group the proportion of CD4+IL-17+ Th17 cells in the model group increased significantly(P<0.05),while the proportion of CD4+CD25+Foxp3+Treg cells decreased(P<0.05);Compared with the model group,the proportion of CD4+IL-17+Th17 cells in the acupuncture group showed a downward trend(P<0.05),while the proportion of CD4+CD25+Foxp3+Treg cells increased(P<0.05);Comparison with blank group,Th17/Treg increased significantly in the model group(P<0.05);compared with the model group,the Th17/Treg in the acupuncture group showed a downward trend(P<0.05).5 Western blot results showed that the expression of Foxp3 protein in the asthma model group was reduced compared with the blank group(P<0.05),and the difference was statistically significant.After acupuncture intervention,Foxp3 protein in the acupuncture group increased significantly(P<0.05),The difference was statistically significant;the expression of STAT3 protein in the asthma model group was significantly increased compared with the blank group(P<0.05).The difference was statistically significant.After acupuncture intervention,the STAT3 protein in the acupuncture group was significantly reduced(P<0.05),the difference is statistically significant.Conclusions 1 The number of leukocytes in asthma model rats is significantly increased,and they are involved in airway inflammation.Acupuncture can significantly reduce the number of inflammatory cells and control airway inflammation.2 The proportion of Th17 cells in monocytes of asthma model rats is increased,and the proportion of Treg cells is decreased.The increase of Th17/Treg indicates that the asthma rats are in an immune imbalance state.The detection of white blood cell count can also support this result.After acupuncture treatment,Th17/Treg showed a downward trend,indicating that the immune imbalance of asthma rats was improved after treatment.3 The expression of STAT3 protein in Lung tissue of asthma model rats was significantly increased,while the expression of FOXP3 protein was significantly reduced.After acupuncture treatment,the expression of STAT3 protein in the acupuncture group was inhibited,and the expression of FOXP3 protein was significantly increased,indicating that acupuncture treatment significantly inhibited the expression of STAT3 protein in the lung tissue of asthmatic rats and promoted the expression of FOXP3 protein,thus regulating the immune imbalance of Th17/Treg.Figure4;Table3;Reference 188...
