| Objective Cell migration is controlled by a variety of proteins,which is the basis of invasion and metastasis of cancer cells.In recent years,Engulfment and cell mobility 1,Rho associated coil containing protein kinase 1 and Rho associated coil containing protein kinase 2 play an important role in cell migration and are related to the invasion and metastasis of lung cancer,liver cancer,gastric cancer and other malignant tumors.In the area of NPC,the study of these proteins is still rare.We detected the expression of Elmo1,Rock1 and Rock2 in nasopharyngeal carcinoma and analyzed the relationship between the expression of Elmo1,Rock1 and Rock2 and the clinicopathological characteristics of nasopharyngeal carcinoma.Then we preliminarily discussed the relationship between the expression of Elmo1,Rock1 and Rock2 and the occurrence,development,invasion and metastasis of nasopharyngeal carcinoma.We expect to provide a new idea for the diagnosis,treatment and prognosis of nasopharyngeal carcinoma.Methods Experimental group: We collected 65 tissues of nasopharyngeal carcinoma from October 2015 to October 2019 in the Department of Otolaryngology and head and neck surgery of Affiliated Hospital of Qingdao University.Control group: We collected 28 Chronic nasopharyngitis specimens.The expression of Elmo1,Rock1 and Rock2 in experimental group and control group were detected by immunohistochemical staining.SPSS 26.0 for Mac statistical software was used to process the data.The expression of Elmo1,Rock1 and Rock2 in experimental group and control group were tested.Then we discussed the relationship between the above proteins and the clinical characteristics of NPC patients.Finally,Kaplan-Meier method was used in survival analysis of nasopharyngeal carcinoma.Results 1.The positive rates of Elmo1,Rock1 and Rock2 in NPC group were higher than those in control group.The differences were statistically significant(χ 2 = 21.343,P <0.05;χ 2 = 19.323,P < 0.05;χ 2 = 9.347,P < 0.05).2.In T-grading,the positive rates of Elmo1 and Rock2 in T3 and T4 were higher than those in T1 and T2.The differences were statistically significant(P < 0.05;χ 2 = 5.149,P < 0.05).The positive rate of Rock1 in T1 and T2 was 71.7%,and the positive rate of T3 and T4 was 78.9%.The difference has no statistical significance(χ 2 = 0.085,P > 0.05).3.In cervical lymph node metastasis,the positive rates of Elmo1,Rock1 and Rock2 in NPC with cervical lymph node metastasis were higher than those without(χ 2 = 6.871,P < 0.05;χ 2 = 6.414,P <0.05;χ 2 = 7.017,P < 0.05).4.In clinical stage,the positive rates of Elmo1,Rock1 and Rock2 in stage III and stage IV were higher than those in stage I and stage II.The differences were statistically significant(χ 2 = 6.904,P < 0.05;χ 2 = 4.392,P < 0.05;χ 2= 25.147,P < 0.05).5.The results of Kaplan-Meier survival analysis showed that the disease-free survival rates of 0.5-year,1-year and 3-year in NPC patients with positive expression of Rock2 were higher than those with negative expression(χ 2 = 6.221,P <0.05).Although the disease-free survival rates of 0.5-year,1-year and 3-year in NPC patients with positive expression of Elmo1 and Rock1 were also higher than those with negative expression,the differences have no statistical significance(χ 2 = 2.969,P > 0.05;χ 2 = 2.888,P > 0.05).6.The positive expressions of Elmo1,Rock1 and Rock2 were not related to the gender(χ 2 = 0.975,P > 0.05;χ 2 = 1.171,P > 0.05;χ 2 = 0.595,P > 0.05)and the age(P > 0.05;χ 2 = 1.789,P > 0.05;χ 2 = 0.176,P > 0.05)of experimental group.Conclusions: 1.Elmo1,Rock1 and Rock2 are positive in NPC.2.Elmo1,Rock1 and Rock2 are related to the cervical lymph node metastasis and clinical stage.Elmo1 and Rock2 are related to the T-grade.3.Rock2 is related to the prognosis.4.Elmo1,Rock1 and Rock2 can be used as tumor biomarkers to observe the invasion and metastasis of NPC. |
