| Objective:Ovarian epithelial neoplasm is the malignant tumor with the highest mortality and lowest survival rate in the gynecological system due to its biological characteristics.The early onset of occult or atypical is not easy to be found.Most of the clinical patients have been treated in the late stage of definite diagnosis,and it is difficult to give effective treatment measures if the best opportunity for treatment is lost.Further to explore the disease development mechanism and seek the more immediate diagnosis or treatment is of great significance to patients.MYOF is a Ca2+dependent membrane phospholipids binding protein belonging to the Ferlin protein family and participating a variety of biological actions involved in cells.A variety of C2 domains,including MYOF,are involved in the biological behavior of both normal and tumor cell.To investigate the expression and clinical significance of MYOF(Myoferlin)in ovarian epithelial carcinoma and its postoperative prognosis base on its characteristic in clinic.Methods:Through dynamic analysis of gene expression profile,TCGA and GTEx data sets were selected to analyze and compare the expression of MYOF gene in normal ovarian tissue and ovarian cancer organization.Immunohistochemical method was used to measure the expression of MYOF in tissues from 55 ovarian epithelial carcinoma,13 borderline,19benign ovarian tumors and 14 normal tissues.Univariate Cox proportional hazard model were statistically assessed to investigate its association with clinicopathological factors and survival rate of ovarian cancer.Results:MYOF was mainly located in the cytomembrane.The level of the expression in ovarian epithelial carcinoma was significantly higher than in borderline,benign ovarian tumors and normal tissues(98.2%,92.3%,31.6%,13.2%respectively)(P<0.05),The high expression rate of MYOF in ovarian malignant tumor was(76.4%),which was also significantly higher than that of epithelial transitional ovarian tumor(23.1%),benign ovarian epithelial tumor(5.3%)and normal ovarian tissue(0%)(P<0.05).MYOF revealed dramatically increased presentation in ovarian epithelial carcinoma with elevatory FIGO stage(P<0.05),MYOF is an independent factor for poor prognosis of ovarian epithelial carcinoma(P<0.05).Conclusion:Our study demonstrate that expression of MYOF was associated with the prognosis of ovarian cancer,it might be a potential prognostic biomarker in OV patients. |
