| Liver fibrosis is a major disease that seriously threatens people’s health and life.It has become an urgent task for medical workers to explore and develop innovative drugs for the prevention and treatment of hepatic fibrosis disease.Traditional Chinese medicine(TCM)believes that the treatment of this disease should focus on the method of"blood softening liver".Based on the theory of traditional Chinese medicine,this study selects the Radix Paeoniae Alba of nourishing blood and softening liver from the classic"Shu Rou Gan"classic prescription,which is combined with the Radix Bupleuri of Liqi Shugan to form a relaxing medicine that has obvious effect on liver fibrosis"Bupleurum-White Peony".In this paper,the pharmacokinetic behaviors of the main components in the blood samples of rats treated with Bupleurum-White Peony and the correlation between endogenous substances,drug components and metabolites were investigated.As to provide a basis for the study on the in vivo effects of Bupleurum-White Peony.The paper is divided into four chapters,mainly from the following three aspects of the study:1.Based on literatures-tracking of these main active ingredients of"liang guanxi"drug pair of Bupleurum-White Peony,this work developed a HPLC-DAD method for determination of these active components including saikosaponin a,saikosaponin c,saikosaponin d,paeoniflorin,albiflorin and oxypaeoniflorin in the extract of Bupleurum-White Peony.We completed quality assessment of the drug pair of the Bupleurum-White Peony,which provide the basis for its metabolic research.2.Using the HPLC-DAD method to observe the changes of exogenous substances and endogenous substances in the extract of Radix Bupleuri and Radix Paeoniae Alba in normal rats through time series,taking into account the correlation between time series and time series.Using SPSS software was used to find the correlation between endogenous substances and endogenous substances,endogenous substances and exogenous substances.The results showed that there were 13 strongly related substances after administration.Among them,there are 12 endogenous substances and 1 exogenous substance.At the same time,combined with ion trap mass spectrometry,the components of saikosaponin a,saikosaponin c,paeoniflorin,albiflorin and oxypaeoniflorin were found in the plasma of rats.3.The LC-MS technique was used to establish a quantitative analysis method of saikosaponin a,saikosaponin c,paeoniflorin and oxypaeoniflorin in rat plasma.The pharmacokinetic behaviors of Bupleurum-White Peony to the plasma of saikosaponin a,paeoniflorin and oxypaeoniflorin in rats were monitored.These results showed that the established method is efficient and simple.From the pharmacokinetic parameters,it can be known that the concentration-time profiles trend of saikosaponin a,paeoniflorin and oxypaeoniflorin after extract of Bupleurum-White Peony is consistent,and the Tmax of the three compounds are similar.All of them have the characteristics of fast absorption and elimination.These pharmacokinetic behaviors help to exert the curative effect in time and achieve the purpose of treating diseases. |
