| ObjectiveTo observe the effect of acupuncture on the expression of 5-HT in the intestinal flora,intestinal mucosal barrier,cerebral tissue in ischemic penumbra of rats with focal cerebral ischemia.Methods90 male Sprague-Dawley rats were randomly divided into sham operation group,model group,acupuncture group,rehabilitation group and acupuncture group,each group was divided into three subgroups of 3d,7d and 14 d,each subgroup have 6 rats.In addition to the sham operation group,the remaining four groups used a modified Koizumi thread embolization method to prepare a permanent cerebral ischemia model.The sham operation group and model group were not treated,the acupuncture group was treated with scalp acupuncture,the rehabilitation group was given treadmill rehabilitation training,acupuncture and rehabilitation group was given both treatments.On the 14 th day after operation,the real-time fluorescence quantitative PCR(q RT-PCR)method was used to detect the changes of intestinal flora(Bifidobacterium,Lactobacillus,Escherichia coli,Enterococcus)of rats in each group.At a predetermined time point,the modified neurological deficit score(m NSS)was used to detect the neurological deficit of each group of rats;the enzyme-linked immunosorbent assay(ELISA)was used to detect serum endotoxin(LPS)and diamine oxidase in the intestinal mucosal barrier index of rats(DAO)expression;Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of 5-HT in the brain tissue of ischemic penumbra.Results1.Modified neurological deficit score(m NSS): Compared with the sham operation group at 3d,7d,and 14 d after operation,the m NSS score increased(P<0.05);the m NSS score decreased in each treatment group compared with the model group(P<0.05);7d and 14 d after operation,the m NSS scores ofthe acupuncture group were significantly lower than those of the acupuncture treatment group and the rehabilitation treatment group(P<0.05).2.The number of target bacteria: 14 days after operation,the number of Lactobacillus and Bifidobacterium decreased,and the number of E.coli and Enterococcus increased in the model group compared with the sham operation group(P<0.05);each treatment group and model group In comparison,the number of Lactobacillus and Bifidobacterium increased,and the number of Escherichia coli and Enterococcus decreased(P<0.05);compared with the acupuncture treatment group and rehabilitation treatment group,the Lactobacillus and Bifidobacterium The number of serotonin increased significantly,and the number of E.coli and enterococci significantly decreased(P<0.05).3.Intestinal mucosal barrier indicators(serum LPS,serum DAO):Serum LPS content: Compared with the sham operation group,the serum LPS content increased in the model group at 3d,7d,and 14 d after operation(P<0.05);the serum LPS content decreased in each treatment group compared with the model group(P<0.05);At 7d and 14 d after operation,compared with acupuncture treatment group and rehabilitation treatment group,the content of serum LPS was significantly reduced in the acupuncture group(P<0.05).Serum DAO content: Compared with the sham operation group,the serum DAO content increased at 3d,7d,and 14 d postoperatively(P<0.05);the serum DAO content decreased at each treatment group compared with the model group(P<0.05)Compared with the acupuncture treatment group and the rehabilitation treatment group,the serum DAO content of the acupuncture group was significantly lower than that of the acupuncture treatment group and the rehabilitation treatment group at 7 and 14 days after operation(P<0.05).4.5-HT content of brain tissue: Compared with the sham operation group,the 5-HT content of brain tissue decreased at 3d,7d,and 14 d postoperatively(P<0.05);compared with the model group,the brain tissue of each treatmentgroup was 5-HT content increased(P<0.05);3d,7d,14 d after operation,compared with acupuncture treatment group and rehabilitation treatment group,5-HT content of brain tissue increased significantly in acupuncture group(P<0.05).Conclusions1.Acupuncture can reduce the score of neurological deficit in rats with cerebral ischemia and promote the recovery of neurological function.2.Acupuncture can increase the number of Lactobacillus and Bifidobacterium in the intestine of cerebral ischemia rats,reduce the number of E.coli and Enterococcus,and optimize the structure of intestinal flora.3.Acupuncture can reduce the content of serum endotoxin and diamine oxidase in cerebral ischemic rats,and promote the repair of intestinal mucosa injury.4.Acupuncture can increase the content of serotonin in cerebral tissue of ischemic rats and promote the recovery of damaged tissues and neurons. |
