Nanotechnology is one of the important means for human beings to understand and study the micro and nano worlds.In particular,the development of nano measurement technology has realized the measurements of size and displacement with nano-scale precision.Scanning electron microscopy and scanning probe microscopy are important branches of nanomeasurement technology.With the rapid development of nanomeasurement technology,it has shown great prospects in the fields such as novel nanomaterials,microelectronics,biomedincine,aerospace,energy and environment.It has shown incomparable advantages in biomedicine.In this thesis,the current research development,imaging characteristics and technical advantages of the two nanomeasurement technologies were summarized.At the same time,atomic force microscope(AFM)and scanning electron microscope(SEM)were used to analyze the surface morphology,physiological characteristics,cell height,cell surface adhesion,and Young’s modulus of various cancer cells.Two cell groups,including liver cells and liver cancer cells,skin cells and skin cancer,were seclected for the experiments,and their ultrastructural changes and mechanical properties were characterized with the treatment of Phellinus igniarius extract.Studies have found that the cancer cell ultrastructures,especially the cell surface adhesion and Young’s modulus,changes significantly before and after the drug treatment.Finally,the parameters of the two nano-measurement technologies were optimized.By adjusting the physical parameters such as the spot value,voltage,and scanning frequency,the optimal conditions for cell measurement were determined.The gray scale analysis and texture feature extraction of cell images were performed by MATLAB,which provided an additional means for comparing and analyzing the differences between cancer cells and normal cells from the cellular physical features.Experiment results have shown that the research and analysis of cancer cells based on nano-imaging technology provide a new method for the studies of related diseases and drugs.This work has important application values for the development of nano-imaging technology in biomedicine.
