Poria cocos Carboxymethyl pachyman(CMP)has a wide range of pharmacological activities.We determined to analyze the respective aggregated configuration of three CMPs distributed in a solvent.By studying the mechanism of three CMP-induced apoptosis of AV3 and HeLa cells,screening for the carboxymethyl pachyman configuration with better tumor cell inhibition.The main findings of this paper are as follows:1.The average molecular weights of the three CMPs group were 4.019×106 Da,1.474×105 Da and 4.373×104 Da respectily.CMP-y1 has a spheroid distribution with a tighter molecular structure and a linear polymer with a tighter molecular structure of CMP-y2 and CMP-g.The molecular composition of CMP-y2 is more abundant than that of CMP-g.2.The results showed that CMP-y1 and CMP-y2 can effectively inhibit the proliferation of AV3 and HeLa cells.While CMP-g can effectively inhibit AV3 cells,ineffective in inhibiting HeLa cells.The inhibition rate of CMP-y1 on AV3 and HeLa cells was higher than that of the other two carboxymethyl pachyman samples(P<0.01).3.The apoptosis rate of the CMP-y1 group reached 92.26 % of the 5-FU group on AV3 cells after 24 h.Compared with the NC group,the apoptosis rate of the CMP-y1,CMP-y2,and 5-FU groups was significantly increased(P<0.01).The apoptosis rate in CMP-y1 group reached 60.27 % vs 5-FU group on HeLa cells after24 h.The apoptosis rates of CMP-y1,CMP-y2 and 5-FU group were significantly increased(P<0.01)compared with NC group on HeLa cell.CMP showed different degrees of inhibitory effect on the two tumor cells,and the inhibitory effect was in a concentration-dependent manner in the concentration range of 75-2400 μg/mL.Among 24 h,the inhibition effect on AV3 cells was the best.The order of inhibition on AV3 and HeLa cells of the three components was CMP-y1> CMP-y2> CMP-g.4.CMP-y1 and CMP-y2 can effectively up-regulate the expression of Caspase-3and Cyto C mRNA and apoptotic proteins in HeLa cell death receptor pathway,and induce HeLa cell apoptosis.CMP-y1 induces apoptosis of AV3 cells,and effectively up-regulates the expression of Bax,p53,Cyto C proapoptotic proteins and mRNAgenes in AV3 cells,and decreases Bcl-2/Bax The ratio in the cells.Mitochondria release a variety of pro-apoptotic factors.It can promote the apoptosis of AV3 cells by activated downstream caspase-8 and caspase-3 protein and up-regulate mRNA gene expression.Our data demonstrate that the spherical CMP-y1 is varied in molecular species,which antitumor activity and comprehensive utilization value are superior to linear carboxymethyl hydrazine polysaccharide.
