| Objective The article aims to study the mechanism of hypoglycemic activity by extracting,purifying and lowering blood sugar on the polysaccharides from branches of Actinidia arguta cut in winter as raw materials.Methods The hot water extraction was used to extract the polysaccharides from the branches of Actinidia arguta.In single factor experiment,the effects on the ratio of liquid-to-material,extraction temperature and extraction time on the extraction rate of the polysaccharides were investigated respectively.Response surface methodology was used to optimize the optimum extraction process of polysaccharides from Actinidia arguta.Deproteinization and depigmentation were carried out on the polysaccharides from the branches of Actinidia arguta,and DEAE-52 cellulose ion exchange chromatography was used to separate the polysaccharides from the branches of Actinidia arguta.The model of mice with Type II diabetic was established by feeding high-fat and high-sugar diet combined with injecting low-dose streptozotocin(STZ)into abdominal cavity.The experimental animals were divided into blank group,model group,low-dose group,middle-dose group,high-dose group and positive group.The body weight and fasting blood glucose of mice were monitored every week.The serum biochemical indexes(MDA,SOD)and four items of blood lipid(TC,TG,HDL-C,LDL-C)were measured.The indexes of pancreas,liver,kidney and other organs were calculated.The liver and pancreas tissue of mice were stained with He in each group and the histomorphology was observed.Results The optimal extraction technology for branches of Actinidia arguta was described as:the ratio of liquid-to-material as 29:1 m L·g-1,the extraction temperature as 80℃,the extraction time as 128 min.The experiment was repeated 3 times,and the average extraction rate was 3.31%.The crude polysaccharides were deproteinized by trichloroacetic acid method from Actinidia arguta,and then decolorized by AB-8 macroporous adsorption resin.DEAE-52cellulose ion-exchange column chromatography was used to separate 4 components of polysaccharides from Actinidia arguta,and one of the major components was studied for hypoglycemic effect.In the hypoglycemic experiment,compared with the model group,there were significant differences in body weight,fasting blood glucose,serum biochemical indicators,and blood lipids of mice(P<0.01)in the AABP3 high-dose group.Compared with the positive group,there was no significant difference(P>0.05).Conclusion Hot water extraction method was used to extract the polysaccharides from the cut branches of Actinidia arguta in this study.The experimental operation method was simple,safe and reliable.Moreover,it has a certain regulating effect on the blood glucose and serum indicators of mice with STZ-induced diabetic,which has great significance for the development and utilization of Actinidia arguta. |
