| Objective:In this study,a comparative analysis of papillary thyroid carcinoma tissues and adjacent tissues was performed using a high performance liquid chromatography-mass spectrometry metabolomics platform.Screening differential metabolites,finding relevant metabolic pathways,exploring potential biomarkers of papillary thyroid and providing new ideas for the diagnosis and treatment of thyroid cancer.Method:A total of 40 thyroid cancer tissue samples from 40 patients with pathologically confirmed thyroid cancer and their paired adjacent tissue samples were collected from the Department of Nail Surgery of Hunan Provincial People’s Hospital from October 2018 to February 2020,a total of 80 cases.HPLC-quadrupole time-of-flight tandem mass spectrometry was used to analyze metabolites in cancer tissue and adjacent tissue samples.Metaboscape 3.0 software was used to preprocess peak metabolomics data,such as peak extraction,denoising,and normalization.In Simca-P14.1 software,principal component analysis(PCA),partial least squares discriminant analysis(PLS-DA),and orthogonal partial least squares discriminant analysis(Orthogonal partial least Square discriminantanalysis(OPLS-DA)and other analysis methods and screening according to variable weight values(VIP> 1)and T test(p<0.05)can characterize the different metabolites between different groups.MetaboAnalysis 4.0 was used for biological analysis of differential metabolites to find abnormal metabolic pathways associated with papillary thyroid carcinoma.Result:1.There are significant differences in metabolic fingerprints between papillary thyroid carcinoma and adjacent tissues.2.Obtaining the original metabolomics data,it was found that there were significant differences in tissue metabolism profiles between the PTC group and the N control group.Combining VIP> 1,FC> 2,and p<0.05,76 potential small molecule differential metabolites were obtained.Among them,33 metabolites showed an upward trend,namely Leucyl-phenylalanine,2-Oxomelatonin,Vanillacticacid,Aspartyphenylalan ine,2-Methylcitric acid;43 metabolites showed a downward trend,respectively:3-glucoside,glycerophospholipid,Phosphatidylcholine,lactose.3.Thirteen abnormal metabolic pathways were detected in this experiment,such as Sulfur metabolic pathway,Taurine and hypotaurine metabolic pathway,Aminoacyl-tRNA biosynthesis,the retinol metabolic pathway,cysteine and methionine metabolic pathways and glycerophospholipid metabolic pathways,pyrimidine metabolicpathways,galactose metabolic pathways,Alanine,aspartate and glutamate metabolic pathways,Glyoxylate and dicarboxylate metabolic pathway,Arginine biosynthesis,Citrate cycle and Pyruvate metabolic pathway.These pathways may be involved in the thyroid pathology of papillary carcinoma.Conclusion:There are significant differences in the metabolic profiles of papillary thyroid carcinoma and adjacent tissues;There are 76 differential metabolites in papillary thyroid carcinoma with different expression levels,such as leucine,2-oxamelatonin,vanillic acid,3-glucoside,glycerophospholipid,phosphatidyl Choline and lactose;glycolysis,amino acid metabolism,choline and phospholipid metabolism are involved in the pathological process of papillary thyroid carcinoma. |
