| Background Periodontitis,caused by local factors,is a common oral inflammatory disease that occurs in periodontal support tissues and can harm the health of the whole body.Chronic kidney disease is a chronic kidney structure and dysfunction caused by a variety of reasons(history of kidney damage is more than 3 months),including a variety of primary and secondary glomerulonephritis,tubular damage and renal vascular lesions,etc.Since renal fibrosis is a common pathological process of most chronic kidney disease to the end stage,renal fibrosis is essential to study the development of chronic kidney disease.Although the correlation between periodontitis and chronic kidney disease has been reported in the literature,so far,the mechanism between the two is still unclear and animal experiments are relatively insufficient.The causes of chronic kidney disease are complex.The establishment of different animal models is very important to explore the relationship between periodontitis and chronic kidney disease.Renal fibrosis is an important part of the progression of various chronic kidney diseases to end-stage renal disease,it’s important to the development of chronic kidney disease.Because there has been no animal experimental research related to periodontitis and renal interstitial fibrosis so far,so this article based on establishing a compound animal model of periodontitis and renal interstitial fibrosis to explore the effect of periodontitis on renal interstitial fibrosis and the mechanism between this two.Purpose To establish a mouse model of periodontitis and renal interstitial fibrosis,then study the effect of periodontitis on renal structure of mice and expression of related factors in renal interstitial fibrosis.Methods 6-week C57 BL / 6 male mice were divided into control group,periodontitis group(PD group),renal interstitial fibrosis group(UUO group),and periodontitis with renal interstitial fibrosis group(PD + UUO group).The periodontitis model was established by silk ligation,and the renal interstitial fibrosis model was established by ligating the unilateral ureter and then cut the ureter.After adaptively feeding mice for one to two weeks,the periodontitis model was established first.The renal interstitial fibrosis model of the mice was established 6 days after the periodontitis model was established,and all the mice were sacrificed 2 weeks after the UUO model was established.The m RNA level of tissues’ fibrosis factors and inflammatory factors were measured by q RT-PCR.The immune cells were analyzed by flow cytometry.And the renal histopathology was observed by HE and Masson staining.Results 1.The UUO group and the PD + UUO group showed obvious changes in kidney morphology on the ligation site,and histological staining showed inflammatory infiltration,renal tubular dilatation,and obvious renal interstitial fibrosis.The PD group and the PD + UUO group showed obvious alveolar bone resorption,and the expression of inflammatory factors in gingival tissue were significantly higher than those in the other two groups(p <0.05).The expression levels of α-SMA and collagen I in kidney tissues were higher in UUO group and PD + UUO group than in the control group and PD group,respectively.2.The expressions of TNF-α,IL-1β,Ly6 G,IL-17 A,and MMP9 m RNA in the kidney tissues of the PD + UUO group were significantly higher than those of the UUO group(p <0.05).There was no significant difference in collagen I m RNA expression(p> 0.05).In addition,the neutrophils in the PD + UUO group were significantly higher than those in the UUO group,and the IL-17 A produced by the T lymphocytes in the PD + UUO group were obviously higher than those in the UUO group.Histopathological staining results showed that there was no significant difference in the degree of inflammatory infiltration and kidney injury in the PD + UUO group compared with the UUO group,and the area of renal fibrosis slightly difference,but the difference was not statistically significant(p> 0.05).Conclusions 1.A mouse model of periodontitis and renal interstitial fibrosis was successfully established.2.In the kidney of renal interstitial fibrosis,periodontitis can up-regulate the expression of TNF-α,IL-1β,IL-17 A and MMP9,promote the infiltration of neutrophils,and aggravate the inflammatory response. |
