Background In recent years,fat transplantation has been increasingly recognized for its many clinical benefits.However,there are inherent defects in transplanting adipose tissue,such as fat liquefaction,necrosis,calcification,etc.The reasons above affect the long-term preservation rate of adipose tissue to a certain extent,and it has become a major problem in the current study of adipose tissue transplantation.Many methods have been lunched on how to improve transplant survival and have achieved good results.Fat transplantation is accompanied by tissue damage inevitably.There is an inflammatory response in the recipient area after transplantation.Kolliphor? P188 is known to be a synthetic surfactant that can repair cell membrane perforations caused by injury,restore cell membranes to normal functions,and maintain basic cell functions.Objective Through the rat experiment of autologous fat transplantation,to explore whether Kolliphor?P188 could inhibit the inflammation during autologous fat transplantation,from the perspective of inflammation to analyze the effect of P188 on transplantation.Methods 36 rats(200-300g)were chosen randomly and raised in SPF lab.To establish the animal models of autologous fat transplantation,which were divided into the control group(Normal saline mixed autogranular adipose tissue transplantation group)and the experimental group(p188 mixed autogranular adipose tissue transplantation group).Weight and volume were measured after fat tissue was removed at time points 1,2and 4weeks.The degree of inflammatory infiltration of fat after transplantation was observed by HE staining.Finally,the m RNA and protein levels of inflammation-related factors IL-1α,1L-1β,IL-6,TNF-α were detected by RT-PCR and ELISA.Determine whether p188 can inhibit the degree of inflammation and infiltration after transplantation of autologous granular adipose tissue in rats,and predict the long-term survival rate.Results There were significant differences in fat weight and volume between the control group and the experimental group(weight: 0.13±0.02 vs 0.19±0.02,P<0.01)at 4weeks.Volume: 0.15±0.03 vs 0.19±0.01,P<0.05);The results of HE staining showed that the inflammatory infiltration area of P188 group was significantly lower than that of normal saline group 4 weeks later.RT-PCR and ELISA results showed that 4 weeks after fat transplantation,P188 significantly lower the m RNA levels of inflammatory cytokines IL-1α,1L-1β,IL-6,TNF-α,and inhibited the protein expressions of IL-1α,1L-1β,IL-6,TNF-α.Conclusion P188 can inhibit inflammatory infiltration after autologous fat transplantation in rats. |
