| Objective:Circular RNA(circRNA)is an endogenous circular loop closed RNA molecule.It is different from long-chain non-coding RNA molecules,and has the characteristics of structural stability,functional diversity,sequence conservation and tissue specificity.We used real-time fluorescence quantitative technology(Real Time-PCR,hereinafter referred to as RT-PCR)to detect the expression of circFOX,circGSE and circLPAR1 in breast cancer and adjacent breast tissues.Statistical analysis of specific expression data results in breast cancer and adjacent breast tissues,to explore its expression in breast cancer and breast adjacent breast tissues,the mechanism of breast cancer development and its clinical significance.Methods:Collect 25 groups of breast cancer and adjacent breast tissue specimens.The Trizol method was used to extract RNA from breast cancer and adjacent breast tissue,reverse transcription was performed on the total RNA extracted,and real-time fluorescence quantification technology was used to detect the expression of circFOX,circGSE and circLPAR1.SPSS 17.0 and Graphpad5.0 were used to process and analyze the experimental data.Spearman rank correlation analysis was performed to analyze the relationship between experimental data and clinicopathological characteristics.Using ROC curve to evaluate the diagnostic value of circFOX,circGSE and circLPAR1 in breast cancer.Results:1.The Δ Ct value of circFOX in breast cancer tissues(13.01±1.46)is higher than the ΔCt value in breast tissues adjacent to cancer(11.45±1.53),the difference is statistically significant(P<0.05),indicating that circFOX is in the breast The expression level in cancer tissue is lower than that in breast tissue adjacent to cancer;2.The ΔCt value of circGSE in breast cancer tissue(3.72±1.38)is higher than that in breast tissue adjacent to cancer(2.72±0.92),the difference is statistically significant(P<0.05),indicating that circGSE is in the breast The expression level in cancer tissue is lower than that in breast tissue adjacent to cancer;3.The ΔCt value of circLPAR1 in breast cancer tissues(10.78 ± 1.52)is higher than the ΔCt value in breast tissues adjacent to cancer(9.90±1.46),the difference is statistically significant(P<0.05),indicating that circLPAR1 is in the breast The expression level in cancer tissue is lower than that in breast tissue adjacent to cancer;4.Using Spearman rank correlation analysis to analyze the relationship between the expression level of circFOX in 25 cases of adjacent breast tissue and KI-67:the expression level of circFOX in adjacent breast tissue was negatively correlated with KI-67,KI-Patients with high expression of 67 molecules(≥14%are high expression of KI-67)have low levels of circFOX expression in the adjacent breast tissue(P<0.05;correlation coefficient r=0.527);5.Using Spearman rank correlation analysis to analyze the relationship between the expression level of circGSE in 25 cases of adjacent breast tissue and the number of gestational times of the patient showed that the expression level of circGSE in the adjacent breast tissue of the cancer had a positive rank correlation with the number of pregnancy and delivery,The higher the number of mothers,the higher the expression level of circGSE in the breast tissue adjacent to the cancer(P<0.05;correlation coefficient r=-0.497);6.Spearman rank correlation was used to analyze the expression level of circLPAR1 in 25 cases of adjacent breast tissues with age,pregnancy time,ER,PR,Her-2,KI-67,P63,CK5/6,P120,EC A,and tumor stage,The relationship between tumor diameter,lymph node metastasis,etc.showed that no grade correlation was found.7.Using ROC curve analysis,it is found that the area under the circFOX curve is 0.761,and the optimal threshold is 0.48.At this time,the sensitivity and specificity are 84%and 64%,respectively;the area under the circGSE curve is 0.745,and the optimal threshold is 0.40.Time sensitivity and specificity were 64%and 76%,respectively.The area under the circLPAR1 curve is 0.666,and its optimal threshold is 0.40.At this time,the sensitivity and specificity are 80%and 60%,respectively.Conclusion:The expression levels of circFOX,circGSE,and circLPAR1 in breast cancer tissues showed a downward trend compared with the expression levels in adjacent breast tissues,indicating that circFOX,circGSE,and circLPAR1 have a certain effect on breast cancer. |
