| Objective:To investigate the ability of CCL21-CCR7 biological axis to influence the proliferation,migration,invasion and apoptosis of hilar cholangiocarci-noma cells in vitro by regulating EMT.Methods:Transfection of pcDNA3.1(+)-CCR 7 plasmid or shRNA of CCR7 to over-express or knock out CCR7 in cholangiocarcin-o ma cell lines QBC939 and HCCC-9810,and stimulate the culture of cholangiocarci-noma cells by adding CCL21 factor.RT-PCR and Western blot were used to detect the expression of CCR7,E-cadherin,vimentin mRNA and protein in each group of cholangiocarcinoma cells;MTT assay,cell scratch,Trans well,Boy den invasion cha-mber and flow cytometry were used to analyze the proliferation,migration,invasion and apoptosis of cholangiocarcinoma cells in vitro.RESULTS:After overexpression of CCR7,the proliferation,migration and invasion of cholangiocarcinoma cells were enhanced,and the apoptotic rate was decreased.After knocking down CCR7,the proli-feration,migration and invasion of cholangiocarcinoma cells were significantly inh-ibited.The rate increases.The differences were statistically significant(P<0.05).Conclusion:The biological axis of CCL21-CCR7 plays a positive regulatory role in the progression of cholangiocarcinoma by regulating EMT. |
