| Breast cancer is one of the most common invasive cancers among women worldwide and the second leading cause of cancer death in women.Tumor markers are chemicals that can be used as markers of cancer in the body.Sensitive and specific tests for tumor markers are commonly used to assess cancer progression,therapeutic efficacy,and recurrence or metastasis.Cancer antigen 15-3(CA15-3)is a member of the MUC1 family.It is widely used as a serum tumor marker for diagnosis,postoperative monitoring,and monitoring of breast cancer patients’ response to treatment,as it involves reliable,available,rapid,inexpensive,simple,and quantitative testing.However,CA15-3 has a poor sensitivity to specificity,especially in the early stages of the disease.CA15-3 is elevated in healthy individuals and in patients with benign disease and lacks specificity for cancer screening and diagnosis,so it is commonly used to monitor recurrence after treatment.Progesterone receptor membrane component 1(PGRMC1)was found to be overexpressed in breast cancer tissues,which predicted a worse diagnosis.PGMRC1 belongs to the membrane-associated progesterone receptor family.Recent data suggested that PGRMC1 could be used as an apoptotic inhibitor to promote breast cancer progression.The expression of PGRMC1 in breast cancer tissues was significantly different from that in normal breast tissues.Increased PGRMC1 expression is associated with an invasive phenotype and poor prognosis in estrogen receptor-positive and negative breast cancers.In vitro studies have shown that PGRMC 1 is important for progesterone-induced tumor proliferation.Therefore,PGRMC 1 might be a new tumor marker.Most markers are not specific to a particular tumor,so one marker is not enough to diagnose a particular cancer.For disease diagnosis and early cancer recognition,simultaneous determination of several tumor markers by a single immunoassay is essential.In this work,we developed a microfluid-based indirect chemiluminescence enzyme immunoassay(MIC)method to detect cancer antigen 15-3(cal 5-3)and progesterone receptor membrane component 1(PGRMC1)for the diagnosis of breast cancer.The linear ranges of C A 15-3 and PGRMC1 were 10 pg/mL-100 ng/mL and 20 pg/mL-358 ng/mL,respectively.The detection limit(LOD)was 3 pg/mL and 5 pg/mL,respectively.CA15-3 and PGRMC1 could be detected simultaneously on one chip.The critical value of PGRMC1 was(25.27±4.26)pg/mL,and the concentration of CA 15-3 was(1.76±0.24)ng/mL.At the same time,we also established the traditional,enzyme-linked immunoassay(ELISA)detection method.The linear range of PGRMC 1 and CA 15-3 was 0.03-1.6 ng/mL and 0.05-10 ng/mL,respectively.The LOD was 10 pg/mL and 0.02 ng/mL.The critical value of PGRMC1 was(38.24±5.32)pg/mL,and the critical concentration of CA15-3 was(1.98±0.46)ng/mL. |
