Objective:Breast cancer is one of the highest incidence of malignant tumoursin women,and seriously endangers the health of human beings.This study is aimed to research ZIC4 gene expression in breast cancer and adjacent normal tissues and explore its clinicopathological and prognostic roles on patients with breast cancer by using immunohistochemistry,western blotting and qRT-PCR.Methods:78 female breast cancer tissue samples underwent modified radical mastectomy in wuxi xishan people’s hospital from January 2010 to December 2012,were selected for immunohistochemistry,and the role of ZIC4 on clinical pathological parameters and survival were analyzed.Western blot and qRT-PCR were performed on 20 pairs of breast cancer tissues with corresponding adjacent normal tissues.Results:Immunohistochemistry showed that the positive expression of ZIC4 in breast tumors was 30.77%and IRS was 6.79±5.22,significantly lower than those in adjacent normal tissues(P<0.001).In addition,the relative protein expression level(0.7923±0.5530)and mRNA level(0.1923±0.2530)of ZIC4 in tumors were also significantly lower than those in para-cancer tissues(P<0.001).In 78 cases of invasive breast cancer,ZIC4 expression was negatively correlated with histological grade and TNM stage(P<0.01).The 5-year overall survival of the ZIC4 overexpression group was significantly higher than that of the low-expression group(HR=0.346,95%CI:0.206-0.771;P=0.009);Univariate survival analysis showed that ZIC4 expression(P=0.024),histological grade(P=0.005),lymph node metastasis(P<0.001)and TNM stage(P=0.001)were all independent factors for overall survival.In the multivariate survival analysis,ZIC4 expression,histological grade,lymph node metastasis and TNM stage were also found significant effect on the overall survival of breast cancer patients.Conclusion:(1)Compared with normal paracancer tissues,the expression level of ZIC4 was significantly reduced in breast cancer tissues.(2)ZIC4 expression was negatively correlated with histological grade and TNM stage,but not significantly correlated with age,tumor diameter,lymph node metastasis,and sex hormone receptor expression.(3)ZIC4 expression level could be used as an independent factor for the overall survival of breast cancer patients and its high expression indicated that breast cancer patients have better clinical prognosis.
