| Objective:Experimental data were used to analyze the effect of aconite injection on JNK expression in chondrocytes of knee osteoarthritis in rabbits,proving that aconite injection has a good therapeutic effect on knee osteoarthritis.Methods:1.32 New Zealand big-eared rabbits were randomly divided into two groups:blank group and model group,blank group(A)with 7 rabbits and model group with 25 rabbits.After anaesthesia with pentobarbital sodium(30mg/kg),the preconfigured 2%w/v papain and0.03mol/L l-cysteine mixture solution were injected into the rabbit knee joint cavity.The administration time was 1,3 and 5 days.The dosage was 0.1ml/kg.A rabbit with large ears was randomly selected from the blank group and the model group for anatomy to determine the success of the model.Then 24 rabbits in the modeling group were randomly divided into model group(B),sodium hyaluronate control group(C),zhengqing fengning control group(D)and aconitum injection treatment group(E),with 6 rabbits in each group.2.After the success of the building of 1,4,7,10 days respectively to the rabbit after the double knee joint cavity injection experimental reagent,sodium hyaluronate group given sodium hyaluronate injection of 0.2 ml,zhengqin Feng Tong Ning group was given zhengqin Feng Tong Ning injection of 0.2 ml,aconite injection group was given aconite injection 0.2ml,the model group was given saline 0.2 ml,don’t give any deal with blank group.After administration,the rabbits were kept for another 1 week,and the experimental rabbits were sacrificed one by one by air embolization.The articular cartilage of the right posterior knee joint of each group was fully exposed.Finally,articular cartilage was collected:cartilage and subchondral tissue of the tibial plateau of the knee were stored in liquid nitrogen.3.Visually observed the appearance of articular cartilage in each group;Microscopic histopathology was performed to compare the changes of articular chondrocytes.The expression of JNK in chondrocytes was detected by immunohistochemistry.The experimental data were processed by spss19.0,and the measurement data were expressed as(x±s).One-way ANOVA was used for comparison between groups,and P<0.05 indicates statisticalsignificance.Results:1.Visible to the naked eye,articular cartilage damage model group compared with blank group was obviously,aconite injection group,the sodium hyaluronate group and zhengqin Feng Tong Ning group compared with model group,articular cartilage surface have no obvious defect and crack,cartilage edge less fat raw osteophyte,articular cartilage is complete,and aconitic injection group was better than the control group..2.HE staining showed that the model set of articular cartilage cells apoptosis in the blank group,aconite injection group,the sodium hyaluronate group and zhengqin Feng Tong Ning group compared with model group,roughly normal cartilage cells,accidentally saw a white foam sample of apoptotic cells,cell cluster,cascading phenomenon is very few,nuclear lobulated and nuclear pyknosis phenomenon is not obvious,and aconitic injection group is better than the control group.3.Immunohistochemical results showed that the JNK expression level of chondrocytes in the model group was significantly higher than that of the blank group,with statistically significant difference between the two groups(P < 0.05);the JNK expression level of chondrocytes in aconitum injection group,sodium hyaluronate group and zhengqing fengtongning group was significantly lower than that of the model group,with statistically significant difference between the two groups(P < 0.05);the JNK expression level of aconitum injection group and the control groups was not significantly different,with no statistically significant difference(P > 0.05).Conclusion:Aconitum injection inhibited the expression of JNK in articular chondrocytes,protected articular cartilage,and had a significant therapeutic effect on KOA.The occurrence of this mechanism may be related to the blocking of MAPK/JNK signaling pathway. |
