NPR-A over-expression cell HEK293 GCAC3,human renal cancer cell ACHN,human lung cancer cell Calu-1,A549,h1792,H1299,h157 and H460,were selected as the experimental subjects.Doxorubicin hydrochloride was used to induce apoptosis and then BNP of different concentrations was used to act on the cells.The proliferation of each cell line was determined by SRB,the sensitivity of each cell line to BNP was studied by ELISA,the expression and change of apoptosis key proteins in each cell was dected by Western blot,thus making a preliminary exploration of the effect of BNP combine Doxorubicin hydrochloride on apoptosis and its possible mechanism.The experiment results are shown as follows:HEK293 GCAC3,ACHN,Calu-1,A549,h1792,H1299,h157 and H460 were all induced apoptosis after 72 hours of treatment with Doxorubicin hydrochloride(concentration between 2.56 ng/mL and 100 ng/mL).BNP of 40 ng/mL or 320 ng/mL significantly inhibited the apoptosis of HEK293 GCAC3 induced by Doxorubicin hydrochloride of 16 ng/mL(P < 0.05),but BNP has no such effect in other cell lines.Compared with other cells,the sensitivity of HEK293 GCAC3 to BNP was significantly higher than other cells(P < 0.05).The effect of BNP that can inhibit the apoptosis induced by Doxorubicin hydrochloride,its mechanism may be related to after BNP binding to its receptor,it can down-regulate the expression level of DR5,inhibit the activation of apoptosis executive factor caspase-3,and finally led to the activity of PARP was affected.
