Objective:In this study,the expression levels of blood glucose,blood lipid,uric acid,tumor necrosis factor-α,glucose-regulated protein(GRP78)in the retina of diabetic rats and the morphological observation on the retina of diabetic rats were detected by characterization observation and experiment,so as to explore the intervention and therapeutic effect of Yuye decoction on diabetic retinopathy and understand the mechanism.The purpose of this study is to provide experimental basis for the clinical selection of the treatment of diabetes mellitus and its complications of diabetic retinopathy.Methods:Thirty-twomale SD rats aged 6-8 weeks were selected.Except 8 rats as normal control group,24 diabetic rats were randomly divided into model group(n = 8),Yuye decoction group(n = 8)and metformin combined with Yuye decoction group(n = 8).All the experimental animals were numbered in the group.The rats in the model group and treatment group were fed with high-fat diet plus 30mg/kg streptozotocin(STZ)in the left lower abdominal cavity for 4 weeks,and the normal group rats were injected with 0.1mol/L p H=4.2 citric acid-sodium citrate buffer solution in the left lower abdominal cavity.After the model was made successfully,the normal group and model group were given 3ml normal saline every day,the Yuye decoction concentrate of 4g/3ml was perfused daily in the Yuye decoction group,and the metformin combined with Yuye decoction group was infused with 1.5ml Yuye decoctionconcentrate and 1.5ml metformin suspension every day at 8: 00 A.M.for 8weeks.The daily mental status,hair,claw nail,tongue color,tail cleanliness and defecation were observed by characterization observation method,blood glucose,total cholesterol,triglyceride and uric acid were measured,tumor necrosis factor-α was measured by ELISA method,retinal morphological changes were observed by HE staining,and the expression of GRP78 protein in retinal tissue of diabetic rats was observed by immunofluorescence.Results:(1)Comparison of the overall situation of rats in each group: the mental and physiological conditions of rats in the normal group were healthy,and the intake,water and urine intake of rats in the model group increased significantly,with weight gain and obesity in the early stage,weight loss and haggard hair in the later stage.The diet,drinking water and urine volume of rats in the Yuye decoction group and metformin combined with Yuye decoction group were significantly better than those in the model group,and the symptoms of polyphagia and polyuria were all improved.there was no significant difference in somatotype,posture,tail characteristics and defecation between the two groups.Although the coat color of rats in metformin combined with Yuye decoction group was not as good as that in normal group,it was brighter and cleaner than that in Yuye decoction group.(2)Comparison of blood glucose in each group: before treatment,there was a significant difference in blood glucose between the normal group and each group(p < 0.01),but there was no significant difference between the model group and the treatment group(p > 0.05).After 8 weeks of oral administration of drugs,the blood glucose values in the Yuye decoction group and metformin combined with Yuye decoction group were higher than those in the model group(p < 0.05)and significantly higher than those in the normal group(p <0.01).(3)Comparison of the levels of total cholesterol and triglyceride in each group: compared with the normal group,there were significant differences in total cholesterol and triglyceride in the model group,and the levels of TC and TG in each treatment group were significantly higher than those in the normal group.Compared with the model group,the levels of TC and TG in each treatment group were significantly lower than those in the model group(p <0.01).(4)Comparison of the level of serum uric acid in each group: compared with the normal group,the level of serum uric acid in the model group was significantly higher than that in the normal group(p < 0.01).The level of serum uric acid in the other groups was significantly higher than that in the normal group(p < 0.01).Compared with the model group,the level of serum uric acid in each treatment group was significantly lower than that in the model group(p < 0.01).(5)Comparison of TNF-α in each group: there was no significant difference in the level of tumor necrosis factor-α among each group(p > 0.05).(6)Morphological observation of HE staining of retina in each group: in normal group,the structure of each layer of retina was intact and the inner limiting membrane was intact and clear.Rats in the model group: the structure of each layer of retina was incomplete and unclear,local loose edema and sparse cells.In the Yuye decoction group,the structure of each layer of the retina was still regular,the thickness of the retina was higher than that of the model group,and the tissue was slightly edema and loosely arranged.Yuye decoction combined with metformin group: the structure of retina was clear,the tissue edema was significantly less than that in the model group,the arrangement was looser,and the shape was still regular.(7)The expression of GRP78 protein in each group: the expression of GRP78 protein in normal group was significantly lower than that in othergroups,the expression of GRP78 protein in model group was significantly higher than that in other groups,and the expression of GRP78 protein in Yuye decoction group and Yuye decoction combined with metformin group was lower than that in model group,and the expression of GRP78 protein in Yuye decoction combined with metformin group was the lowest.Conclusions:1.The diabetic rat model can be effectively established by feeding high-fat and high-sugar diet combined with streptozotocin injection.2.Metformin and Yuye decoction can effectively improve the symptoms of diabetes,reduce blood sugar,blood lipids and other biochemical indicators,of which the combination of Yuye decoction and metformin is the best.3.Yuye decoction may interfere with the formation and development of diabetic retinopathy by relieving endoplasmic reticulum stress and reducing the expression of GRP78 in the retina of diabetic rats,so as to play a role in prevention and treatment.
