Objectives:Ovarian cancer(OC)is the deadliest cancer among gynaecological malignancies,which lacks effective early diagnosis indicators,and has a high recurrence rate and poor prognosis.Long noncoding RNAs(lncRNAs)have been reported to play a key role in the development and progression of human malignancies.FLJ33360 is an lncRNA with unknown functions.This study was designed to determine the clinical significance and mechanism of FLJ33360 in OC.Methods:The online databases Gene Expression Profiling Interactive Analysis(GEPIA)and The Cancer Genome Atlas(TCGA)were used to explore the expression and clinical significance of FLJ33360 in OC.A total of 32OC specimens and 31 normal ovarian specimens were collected from the Department of Obstetrics and Gynecology in People’s Hospital of Zhengzhou University between Juna 2018 and October 2018.Cancer specimens were collected from OC patients undergoing cytoreductive surgery,and healthy ovarian tissue was collected from patients undergoing adnexectomy due to myoma or uterine prolapse.The expression of FLJ33360 was detected by quantitative reverse transcription polymerase chain reactions(qRT-PCR),and its relationship with clinicopathological parameters of OC patients was analyzed.The potential diagnostic value of FLJ33360 in OC was evaluated using the receiver operating characteristic(ROC)curve.Bioinformatics was used to predict microRNAs(miRNAs)interacting with FLJ33360,extracting data on the expression of related miRNAs in Gene Expression Omnibus(GEO)and ArrayExpress databases,and verifying their expression in OC by meta-analysis.TCGA database and bioinformatics were used to predict the downstream target genes of miRNAs,and Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses of target genes were performed using the GluGO plug-in of Cytoscape 3.6.1.A protein-protein interaction(PPI)network of the target genes was constructed using STRING 10.5.Central target genes were verified using Pearson correlation analysis.Results:The expression level of FLJ33360 in OC tissues was significantly lower than that in normal ovarian tissues(P=0.0011).And the expression level of FLJ33360 was significantly lower in OC patients with stage Ⅲ/Ⅳ compared with patients with stage I/II(P=0.027);the expression level of FLJ33360 was significantly lower in high-grade serous carcinoma compared with patients with low-grade serous carcinoma(P=0.027).And FLJ33360 has potential diagnostic value in OC(area under the curve was 0.793,P<0.001).Bioinformatics and meta-analysis showed that FLJ33360 could competitive bind with miR-30b-3p,and miR-30b-3p was significantly up-regulated in OC.The target genes of miR-30b-3p was mainly involved in the positive regulation of smooth muscle cell migration,the metabolic process of unsaturated fatty acids and the positive regulation of epithelial to mesenchymal transition.The PPI network revealed that BCL2 was a central target gene,and BCL2 was positively correlated with FLJ33360 in EOC(r=0.134,P=0.014).Conclusion:1.The expression level of lncRNA FLJ33360 is significantly lower in OC than normal ovarian tissue;2.FLJ33360 is a potential biomarker for early diagnosis in OC,and is closely related to disease stage and pathological type;3,FLJ33360 can regulate the expression of genes such as BCL2 by sponging miR-30b-3p and thus participates in the development of OC.
