Objective:To observe the metabolic pathways of Short chain fatty acids(SCFAs)and Total bile acid(TBA),investigated the mechanism of improving on glycolipid metabolism by milk-born composite probiotics.Methods:Type 2 diabetic rats were induced by an administration of high-fat diet and an injection of STZ.T2D rats were divided into model group,Metformin group(0.2g/kg),high dose composite probiotics group(1×10~8 CFU/d lactobacillus+1×10~6 CFU/d saccharomycetes),low dose group(1×10~100 CFU/d lactobacillus+1×10~8 CFU/d saccharomycetes),treated for 6 weeks.1.Fasting blood glucose(FBG),glucose tolerance(OGTT),glycated hemoglobin(HbA1c),fasting insulin(FINS),insulin resistance index(IRI),C peptide(CP)and blood lipid(TC,TG,LDL-C,HDL-C)were detected;the morphology of liver and pancreas was observed by HE.2.The contents of glucagon like peptid-1(GLP-1)and peptid YY(PYY)were determined by ELISA;the content of short chain fatty acids(SCFAs)was determined by GC;the expressions of GLP-1,PYY,GPR43,GPR41 mRNA were determined by RT-PCR;the expression of GLP-1 protein was detected by WB.3.The content of TBA was determined by ELISA;the expression of CYP7A1,FXR,TGR5 mRNA were determined by RT-PCR.Results:1.Composite probiotics decreased FBG,OGTT,HbA1c,FINS,IRI,and increase CP levels(all P<0.01).2.Composite probiotics significantly decreased the TC,TG,LDL-C and TBA in serum,increased TBA in liver and feces(all P<0.05),and improved pancreatic and liver tissue lesions.3.Composite probiotics increased the content of SCFAs,GLP-1 and PYY,up-regulated the expression of GLP-1,PYY,GPR43,GPR41mRNA and GLP-1 protein(all P<0.01).4.Composite probiotics up regulated the expression of CYP7A1、TGR5、FXR mRNA(all P<0.05).Conclusion:Composite probiotics improved glycolipid metabolism in T2D rats,the mechanism is related to the regulation of SCFAs-GPRs-GLP-1 and TBA-TGR5-GLP-1 signaling pathways. |
