ObjectiveIn this study,lycium barbarum polysaccharide(LBP)was used in combination with bone marrow mesenchymal stem cells(BMSC)transplantation to intervene the thin endometrium formed by artificial molding.After the intervention,it’s repair function,endometrial thickness improvement effect and endometrium receptive corresponding indexes can be tested.And then its mechanism of action of promoting wound repair will be discussed.Through this research,it is supposed that diet regulating theory of TCM and clinical nursing health education can provide a source of scientific guidance,for medicine edible chemicals in women in the field of assisted reproduction theory provide innovative research ideas and methods.Methods35 rats were put into 5 groups randomly:control group,model group,BMSC group,LBP group and combined group,7 rats in each group.In addition to the control group using normal saline modeling,the other 4 groups were all using 95%ethanol intrauterine injection to establish thin endometrium model.In addition to intrauterine injection of BMSC transplantation solution in the BMSC group and the combination group,all the other groups were injected with intrauterine saline after modeling.In addition to LBP and the combination group,all the other groups were given intrauterine saline injection.After 14 days of intervention,subjects were observed after two estrous cycles.HE staining was used to detect the changes of endometrial tissue structure,thickness,number of blood vessels and number of glands in each group.The expression of endometrial vimentin,keratin and ITG α v β 3 was detected by immunohistochemistry.Protein expressions of VEGF,LIF and ETA in rat endometrium were detected by western-blotting.ResultsModel building:The thin endometrium was successfully built.Morphologically,the endometrium structure of the control group was intact and that of the model group was intact Endometrium thinned,endometrium was less continuous,folds were reduced,part of glandular epithelium was shed,glandular epithelium and endometrium epithelium cells were low columnar,even flat.The stroma showed fibrous hyperplasia,the glands were sparse and some glandular epithelium atrophy.The mean intimal thickness of the model group was thinner than that of the control group(P<0.05).Cells result:Proficient in BMSC culture,identification,counting and subculture techniques and ETA record and observation in the process.Finally,it was digested by trypsin and transplanted into the uterus of rats.HE staining results:compared with the control group,the intima of the model group was thinner(P<0.05)than that of the model group.Endometrial thickness was observed in BMSC group,LBP group and combination group(P<0.05).Compared with the control group,the number of blood vessels in the endometrium decreased in the model group(P<0.05),and increased in the BMSC group,LBP group and the combined group(P<0.05).Compared with the control group,the number of glands in the model group decreased,while that in the BMSC group,LBP group and the combined group increased(P<0.05).Immunohistochemical results:compared with the control group,the endometrial vimentin and keratin of the model group.The mean optical density(MOD)of ITGαvβ3 decreased(P<0.05)and the MOD values of vimentin,keratin and ITGαvβ3 increased(P<0.05)in BMSC group,LBP group and combination group compared with the model group.Western blot results:VEGF,LIF and ETA in the endometrium of the model group were compared with those of the control group.Compared with the model group,MOD values of BMSC group,lycium barbarum polysaccharide group and combination group were increased(P<0.05).ConclusionIn this experiment,rats were injected with 95%ethanol in uterine cavity,which could reduce the thickness of endometrium,disrupt the continuity and reduce the number of folds.The thin endometrium rat model conforming to clinical phenotypic characteristics was successfully established.After identification,countation of BMSC,the quantification and the qualification of the cell is proved enough.LBP gavage combined with BMSC transplantation was more effective than either group in increasing the thickness of thin endometrium and improving the endometrial receptivity of thin endometrium in rats.The results of the study is helpful to promote Chinese wolfberry as the medicine food homology secondary development and application of traditional Chinese medicine,expand the research space,and broad application prospect,promote Chinese wolfberry similar or other substances,diet,medicine edible medicinal food fang in solving clinical problems in further study,in the human life and health play a bigger role.And it is important to make the health diet education more scientific,credible and advanced.In this account,patients can receive professional knownledge to improve the success rate of pregnancy patients.
