Research On The Protective Effect Of Acupuncture Preconditioning On The Activation Of JAK/STAT Pathway In Ischemic Reperfusion Injury

Objective:To research of acupuncture and moxibustion pre processing activation of Janus kinase(JAK)signal transduction and transcription activation factor(STAT)pathway(JAK / STAT pathway)which plays an important role in the mechanism of the protective effect of ischemic reperfusion injury.Methods:Forty male SD rats,which were established to myocardial ischemia reperfusion injury model,were randomly divided to four groups: sham group,model group,acupuncture and moxibustion preconditioning group,AG490 and acupuncture and moxibustion preconditioning group.The rats in the acupuncture and moxibustion preconditioning group were given acupuncture for 7 d before the modeling additionally(each time for 20 min,once a day).At the end of reperfusion injury,the left heart function indexis were detected by HR: heart rate(HR),left ventricular end systolic pressure LVESP,left ventricular diastolic end pressure(LVEDP),maximum and minimum change rate(DP/DT)in each group of rats,the myocardial injury of hearts were detected and observed by TUNEL staining.The protein immunoblotting(Western blot)verification of acupuncture and moxibustion preconditioning or AG490 combined with acupuncture pre treatment effect on the activation of JAK/STAT pathway and the expression of high mobility group protein factor(HMGB1).Results: compared with the model group,left ventricular function of the rats in acupuncture and moxibustion preconditioning group were improved due to the ischemic reperfusion injury.Compared with Acupuncture Pretreatment group,the LVESP of AG490 and acupuncture and moxibustion preconditioning group were decreased 26.6%,LVEDP rose 47.6%,dp/dt Max and dp/dt min have declined,the difference were statistically significant(P<0.05);TUNEL staining indicated that myocardial cell apoptosis index of AG490+of Acupuncture Pretreatment group was 23.7%+2.5%,which compared with the acupuncture preconditioning group was also statistically significant(P<0.05);Western blot proved that acupuncture and moxibustion preconditioning group effectively inhibited the release of HMGB1 and activation of the JAK/STAT pathway.While in the AG490 and acupuncture and moxibustion preconditioning group,the JAK/STAT pathway is blocked,the release of HMGB1 increased(P<0.05).Conclusion: Acupuncture Preconditioning protected the injury of ischemic reperfusion injury by inhibiting the release of HMGB1,the mechanism may be related to the activation of JAK/STAT pathway.
