ObjectiveThrough biochemical and molecular identificationof the isolated strains,and a variety of molecular typing techniques,this study was conducted to better understand drug susceptibility characteristics of five kinds of bacteria obtained from inpatient.Furthermore,to investigate the correlation of different genotypes of five kinds of bacterias with antibiotic resistance,and to propose the theory reference for clinical rational use of antibiotics for treating different genotypes of multiple drug-resistant bacterias.Reach MethodsUsing VITEK 2 Compact automatic microbial system,and gram staining technique,sugar fermentation tests,indole(Imdole)reaction experiment,VP experiment,plasma coagulase test,a number of samples were analyzed by bacterial identification and susceptibility test.We selected clinical commonly used in the treatment of E.coli,klebsiella pneumoniae,staphylococcus aureus,pseudomonas aeruginosa and acinetobacter baumannii infection such as antibiotics [penicillin(PEN),Oxacillin(OXA),Amoxicillin(AMX),Piperacillin(PIP),Cefazolin(CFZ),Cefuroxime(CXM),Ceftazidime(CAZ),Cefepime(FEP),Merope nem(MEM),Moxalactam(MOX),Azithromycin(AZM),Clindamycin(CLI),Vancomycin(VAN),Gentamicin(GEN),Amikacin(AK),Tetracycline(TET),Minoc ycline(MNO),Chloramphenicol(CHL),Ciprofloxacin(CIP),Sulfathiazole(ST),Nitrofura ntion(AHD).Designing specific primers for the different genes of the multiple drug-resistant bacterias and the primer specificity was performed in order to avoid the complementary sequence and the hairpin structure at the 3 ’end.Polymerase chain reaction(PCR)was performed to detect the TEM1,CTX M,KPC,Staphy,NDM,SHV,CMY,DHA,VIM,OXA1,and OXA2 genes.20μL product and 2μL bromophenol blue were mixed in 2% agarose gel electrophoresis by ethidium bromide staining.After 5min,DNA bands were observed by UV Gel imaging system.Results319 strains were successed separation and identification from cooperation hospitals,and the separation rate in the top five E.coli,acinetobacter baumannii,staphylococcus aureus,pseudomonas aeruginosa and klebsiella pneumoniae.The results of antibiotic resistance of five kinds of bacterias with antibiotic resistance.The results showed that the resistance rates of E.coli to PEN,OXA,CFZ,CLI,VAN,GEN,TET,MNO were 76.5% ~94.1%,and the resistance rates to MOX and AHD were about 28.0%.The resistance rate of acinetobacter baumannii to PEN,OXA,CFZ,CLI,VAN,GEN,TET and AHD were 66.0%~91.5%,and the resistance rates to CXM and AZM were about 33.0%.The resistance rate of staphylococcus aureus to MNO,CHL and AHD were 31.5%~47.9%,other 18 kinds of antibiotics were higher resistance(≥56.2%).The resistance rate of pseudomonas aeruginosa to PEN,OXA,AMX,CFZ,CXM,CLI,VAN,MNO and CIP were 79.4%~98.4%,and the resistance rates to MOX and ST were 22.2% ~ 30.2%.The resistance rate of Klebsiella pneumoniae to PEN,OXA,AMX,CLI,VAN,GEN,TET,MNO and ST were 67.6% ~ 95.6%,and the resistance rates to FEP,MEM,MOX and AZM were 22.1% ~ 26.5%.The strains were successed separation and identification from clinicalwere carrying TEM1,CTX M,KPC,Staphy,NDM,SHV,CMY,DHA,VIM,OXA1,OXA2 genes respectively.ConclusionThe drug resistance of five kinds of bacteria obtained from inpatient is serious,and the result of the drug-sensitive is associated with the gentype of the isoaltes.the phenomenon of multiple resistance is very common.The sensitive rate and drug resistance rate of multiple drug-resistant bacterias on the different antibiotics were significantly different,and the resistant mechanism of multiple drug-resistant bacterias to different antibiotics are closely related to TEM1,CTX M,KPC,Staphy,NDM,SHV,CMY,DHA,VIM,OXA1 and OXA2 genes.This study focuses on the severity of bacterial resistance,and aims to provide a theoretical basis for novle drug targets,and provided a strong theoretical basis for people seek to control the multiple drug-resistant bacterias infection.
