Objectives:High-throughput RNA sequencing(RNA-seq)was used to detect long-chain non-coding RNA(lncRNA)gene expression profiles in adult degenerative scoliosis(ADS),lumbar disc herniation(LDH),and healthy adults(normal).To analyze the differential gene expression profiles of lncRNA in adult degenerative scoliosis,and to explore the possible differentially expressed genes and related molecular functions and biosignal pathways of lncRNA in adult degenerative scoliosis to further study adult degenerative scoliosis.The development mechanism and the development of specific molecular biomarkers and precise targeted therapy provide a theoretical basis.Methods:1.High-throughput RNA sequencing(RNA-Seq)was used to detect IncRNA gene expression profiles in 9 blood samples from the ADS group(n=3),the LDH group(n=3),and the normal group(n).The test results of the above three groups of samples were compared two by two.Then,the Wien plot was used to analyze the comparison between the three groups and the intersection of the two ADS vs Normal and ADS vs LDH comparison results to screen for differentially expressed genes commonly found in ADS.Describe the differential gene expression profiles of ADS and ultimately screen for possible differentially expressed genes in ADS.2.Carrying out further biological information analysis on the selected ADS differentially expressed genes,and performing functional enrichment analysis on differentially expressed genes by using GO molecular function analysis software to explore the molecular functions mainly involved in differentially expressed genes;KEGG database signaling pathway analysis to find biological signaling pathways associated with these differentially expressed genes.Results:1.Compared with LDH group,there were 234 differentially expressed genes in IncRNA,including 153 up-regulated genes and 82 down-regulated genes.Compared with Normal group,there were 241 differentially expressed genes in lncRNA,including 153 up-regulated genes.89 genes were down-regulated;compared with the Normal group,there were 212 differentially expressed genes in lncRNA,including 122 up-regulated genes and 90 down-regulated genes.Comparing the differentially expressed genes identified by the three comparison groups,the differentially expressed genes were analyzed.The three groups were compared,and there were 4 differentially expressed genes of lncRNA;ADSvsNormal and ADSvsLDH were compared.In comparison,there were 79 differentially expressed genes in the common lncRNA.2.According to the results of GO enrichment analysis in this study,it is found that in biological processes,most of ADS differentially expressed genes are involved in cellular metabolism,organic matter metabolism,cellular nitrogen compound metabolism,cellular macromolecular metabolism,etc.Related biological processes.In terms of molecular function,most of the differentially expressed genes of ADS are related to protein binding,binding of organic cyclic compounds,and binding of heterocyclic compounds to molecular functions related to binding.In terms of cellular components,a significant amount of differentially expressed genes of ADS are associated with intracellular components,intracellular cytoplasm,intracellular lipids,membrane organelles,and the like.Then,using the KEGG database search,it was found that ADS lncRNA differentially expressed genes involved 20 pathways,such as lysosome,HIF-1 signaling pathway,ubiquitin-mediated proteolysis,Adherens junction,phagosome,mTOR signaling pathway,citric acid cycle.(citrate cycle),heterologous end-joining,ribosome,primary immunodeficiency,B cell receptor signaling pathway,T cell receptor signaling pathway,glycosaminoglycan degradation,viral carcinogenesis,etc.,mostly with inflammation Pre-cytokine signaling is involved;the most prominent pathway for enrichment is lysosomes.Conclusions:1.Adult degenerative scoliosis and lumbar disc herniation、healthy adults,there are many distinct differentially expressed genes such as ENSG00000253352.9,XLOC000500,XLOC135387,etc.,and the difference is most significant with lncRNA XLOC 000209.IncRNA XLOC 000209 may be a key differentially expressed gene of ADS..These differentially expressed genes may be involved in the regulation of the development of adult degenerative scoliosis,providing an important biological basis and reference for the search for molecular markers of adult degenerative scoliosis or the treatment of new targets.2.The functions of these differentially expressed genes are roughly classified into various biological processes such as cell metabolism,protein binding,organic cyclic compound binding,intracellular components,membrane organelles,etc.,and involve multiple signals related to cell synthesis and catabolism and inflammatory reactions.Pathways,such as lysosomes,HIF-1 signaling pathway,B cell receptor signaling pathway,T cell receptor signaling pathway,mTOR signaling pathway,etc.
