The Effects Of High Expression Of ALDH1 In Gastric Tumor Cells Escaping The Killing By M_φ | | Posted on:2021-02-03 | Degree:Master | Type:Thesis | | Country:China | Candidate:G Q Zhang | Full Text:PDF | | GTID:2404330602496042 | Subject:Clinical laboratory diagnostics | | Abstract/Summary: | PDF Full Text Request | | Background:In China,the morbidity and mortality of gastric cancer are significantly higher than the world average due to the lack of specific diagnostic methods and effective treatment in the early stage.Therefore,further identification of gastric cancer and its related regulatory role of biomolecules has important practical significance and clinical value.Recent studies has shown that macrophages polarized toward the M1 type of effector molecules such as tumor necrosis factor-ɑcan kill gastric cancer cells,but high expression of ALDH1 in gastric cancer cells can promote the peritoneal metastasis of cancer cells.We speculate thatALDH1 may play an important role in the attack of gastric cancer cells escaping macrophages.But the exact mechanism is unclear.Objective:1.To investigate the effect of ALDH1 expression on proliferation and invasion of gastric cancer cells.2.Whether high expression of ALDH1 plays an important role in killing gastric cancer cells by escaping from gastric macrophages.Methods:1.The ascites samples of 72 patients were collected and the content of ALDH1 was detected.2.The ALDH1~highigh gastric cancer cell line(SGC-7901)was obtained by serum-free culture and ALDH1~lowow gastric cancer cell line by transfection,and the difference in proliferation and invasion capacity of gastric cancer cells between the two groups was detected.3.Macrophages and its effect on molecular TNF-ɑrespectively with two groups of gastric cancer cell lines to cultivate,and detect whether relevant protein expression level analysis ALDH1 play an important role in killing gastric cancer cells escape gastric macrophages.Results:1.The level of ALDH1 in the ascites of patients with malignant tumor was significantly higher than that of patients with benign disease,and the content of ALDH1 in the ascites of patients with gastrointestinal malignancywas significantly higher than that of patients with gynecologic malignancy.2.The proliferation and invasion capacity of ALDH1~highigh gastric cancer cells were significantly higher than that of ALDH1~lowow gastric cancer cells,and the killing efficiency of macrophages against ALDH1~lowow gastric cancer cells was significantly higher than that of ALDH1~highigh gastric cancer cells.3.The killing effect of M1 type of macrophage effector molecule TNF-ɑon ALDH1~lowow gastric cancer cells is stronger than that of ALDH1~highigh gastric cancer cells,and matrix metalloproteinases MMP2、MMP7 and MMP9 in ALDH1~highigh expression quantity in gastric cancer cells were higher than ALDH1~lowow gastric cancer cells.Conclusion:1.The detection of ALDH1 concentration in ascites has important clinical value in the identification of the source of ascites and assisting in the diagnosis of malignant tumors.2.The gastric cancer cells with high expression of ALDH1 have stronger proliferation and invasion capacity.3.The high expression of ALDH1 gastric cancer cells can escape the killing effect of macrophages by antagonizing TNF-ɑand other effector molecules secreted by macrophages,which is beneficial to the invasion of gastric tumor cells. | | Keywords/Search Tags: | gastric cancer stem cells, acetaldehyde dehydrogenase 1, macrophages, tumor necrosis factor-ɑ | PDF Full Text Request | Related items |
